马铃薯优良品种大西洋、云薯201茎尖诱导技术

【摘要】 【目的】掌握马铃薯优良品种大西洋、云薯201茎尖诱导技术,为马铃薯工厂化脱毒快繁提供技术参考。【方法】以马铃薯优良品种大西洋、云薯201的茎尖作为试验材料,从外植体的消毒灭菌、不同基本培养基、植物激素种类及浓度配比探讨对马铃薯茎尖诱导效果的影响。【结果】在两因素和3因素灭菌剂组合中,以75%酒精30s+10%NaClO5min+0.1%HgCl25min为外植体芽灭菌最佳组合;MS基本培养基比其他基本培养基更利于茎芽的诱导;在茎芽诱导培养基中添加0.02mg/LGA3可以提高芽的诱导率,以6-BA0.5mg/L+NAA0.01mg/L+GA30.02mg/L激素组合处理的效果最好。【结论】建立了适合马铃薯品种大西洋和云薯201茎尖诱导的体系,为其工厂化育苗奠定基础。

【Abstract】 [Objective]This research aimed to master the stem apex induction technology for excellent potato varieties Daxiyang and Yunshu 201 in order to provide technical reference for the virus-free and rapid propagation of factory potatoes. [Method]The stem apex of excellent potato varieties Daxiyang and Yunshu 201 were used as test subjects, then the influential effects of the sterilization, different basic culture media, plant hormone type, and concentration ratio on the stem apex induction of potato were explored. [Result]Among different sterilant combinations of 2 factors and 3 factors, the optimum combination of buds sterilization was using 75% alcohol 30 s+10% NaClO 5 min+0.1% HgCl2 for 5 min. The basic MS culture medium was better than other basic culture media for bud induction. In the culture medium induced bud, adding 0.02 mg/L GA3 could improve the bud induction rate, and the optimum hormone treatment combination was 6-BA 0.5 mg/L+NAA 0.01 mg/L+GA3 0.02 mg/L. [Conclusion]Suitable stem apex induction system for potato varieties Daxiyang and Yunshu 201 was constructed to set the foundation for industrialized potato seedlings.
