DRD3基因敲除鼠饲养繁殖及基因型鉴定
Breeding Reproduction of DRD3 Gene Knockout Mice and Its Genotype Identification
【摘要】 [目的]对于DRD3基因敲除小鼠的饲养繁殖以及鉴定方法进行分析,为进一步研究多巴胺及其受体的功能提供动物模型。[方法]引进的DRD3基因敲除小鼠均为杂合子基因型,1雌1雄同窝进行饲养繁殖,从仔鼠中提取基因组DNA,进行PCR鉴定。[结果]成功获得的子代小鼠有敲除杂合子、野生型及敲除纯合子3种基因型。DRD3敲除杂合子基因型小鼠的饲养繁殖获得成功,亦获得较多敲除纯合子基因型小鼠。[结论]正确的饲养繁殖管理及基因型鉴定方法可从DRD3基因杂合子小鼠成功获得纯合子小鼠。
【Abstract】 [Objective] To discuss the breeding and identification methods of DRD3gene knockout mice and lay the foundation for further study on the function of dopamine and its receptors. [Method]Heterozygote mice were bred and reproduced. Genome DNA extracted from the mice’s tails were amplified by using PCR method for genotype identification. There were three genotypes (heterozygote,wild-type and homozygote) in the offspring. [Result]The experimental model for DRD3gene knockout mice was successfully established and more DRD3knockout mice were reproduced. [Conclusion]Appropriate methods of breeding,reproducing,and identifying are effective ways to gain DRD3gene knock-out mice from heterozygote mice.
