【摘要】 目的优化重组人粒细胞集落刺激因子,以获得工程菌的高密度发酵和目的蛋白的高表达。方法借助正交实验,在14L发酵罐中,研究重组人粒细胞集落刺激因子工程菌在添加不同浓度钾盐、钠盐和镁盐后对质粒稳定性和目的蛋白表达的影响。结果在高浓度盐的环境下,菌株的比生长速率和质粒稳定性均显著提高,从而提高目的蛋白的表达量。结论获得一种高效生产人粒细胞集落刺激因子的方法。

【Abstract】 Objective To optimize the fermentation procedure of recombinant human granulocyte colony-stimulating factor and to obtain high cell density and high level expression of the aim protein.Methods With orthogonal experiment,study the influence of different concentration potassium,sodium salt and magnesium salt on plasmid stability and expression of aim protein in a 14L fermentor.Results Under high concentrations of salt in the culture,the specific growth rate of strains and plasmid stability were significantly improved,so as to improve the purpose of protein expression.Conclusion Gain a efficient fermentation method to produce recombinant human granulocyte colony-stimulating factor.
