补心软脉颗粒的提取纯化工艺优选
Optimization of Extraction and Purification Technology for Buxin Ruanmai Granule
【摘要】 目的:优选补心软脉颗粒的提取纯化工艺。方法:以红景天苷转移率和浸膏得率为综合评价指标,选取加水量、提取时间和提取次数为考察因素,采用正交试验法优选补心软脉颗粒中红景天等5味药材饮片的水提取工艺;以总黄酮醇苷转移率为指标,采用单因素试验考察银杏叶的醇提工艺;以红景天苷的转移率和除杂率为指标,采用单因素试验考察补心软脉颗粒水提液的醇沉工艺。结果:优选的水提工艺为红景天等5味加8倍量水提取3次,每次0.5 h;优选的醇提工艺为银杏叶加10倍量70%乙醇提取2次,每次1 h;优选的醇沉工艺为水提液浓缩至相对密度1.10~1.15,缓缓加入95%乙醇至含醇量70%,边加边搅拌,冷处静置24 h。结论:该优选的提取纯化工艺简便易行,为补心软脉颗粒的大生产提供实验依据。
【Abstract】 Objective:To optimize extraction and purification technology of Buxin Ruanmai granule.Method:With transfer rate of salidroside and yield of extract as comprehensive evaluation indexes,the amount of water,extraction time and extraction times were chosen as investigated factors,orthogonal design was used to optimize water extraction process of Rhodiola crenulata and four other Chinese herbal pieces from Buxin Ruanmai granule;With transfer rate of total flavonol glycoside as index,ethanol extraction technology of Ginkgo biloba piece was investigated by single factor test;With transfer rate and cleaning rate of salidroside as indexes,alcohol precipitation technology of water extraction liquid from Buxin Ruanmai granule was optimized by single factor test.Result:Optimized water extraction technology of R.crenulata and four other Chinese herbal pieces was:extracted 3 times with 8 times the amount of water,0.5 h each time;Optimized alcohol extraction technology was:extracted 2 times with 10 times the amount of 70% ethanol,1 h per time;Optimal alcohol precipitation conditions were as follows:water extraction liquid concentrated to its relative density of 1.10-1.15,slowly added 95% ethanol until the concentration of ethanol of 70%,stirring at the same time,precipitated 24 h in cold.Conclusion:This optimized technology was simple,it could provide experimental basis for large-scale production of Buxin Ruanmai granule.
