β-环糊精衍生物流动相添加剂应用于HPCE中分离布洛芬
Separation of Ibuprofen in HPCE with β-Cyclodextrin Derivative
【摘要】 以双-[-6-氧-(-2-间羧基苯磺酰基-丁二酸-1,4-单酯-4-)-]-β-环糊精(β-CD-B2)为手性添加剂在高效毛细管电泳(HPCE)中分离布洛芬,考察了缓冲溶液硼砂的浓度、pH值,手性添加剂β-CD-B2的浓度,分离电压对布洛芬手性异构体分离的影响.结果8min内实现了对映体的基线分离,最佳分离条件下分离度Rs为4.26.布洛芬对映体分离的前峰线性范围为0.0538~0.942g/L,后峰线性范围为0.178~0.942g/L.在分离布洛芬的最佳条件下,可同时分离布比卡因和布洛芬2种手性药物的混合溶液.
【Abstract】 Chiral separation of ibuprofen was investigated by bis-[-6-oxygen-(-2-m-carboxyl phenylsulfonyl-succinic acid 1,4-ester-4-)-]-β-cyclodextrin(β-CD-B2) as chiral additive in high performance capillary electrophoresis(HPCE).The effect of concentration and pH of borax buffer solution,concentration of chiral additive β-CD-B2 and separation voltage on the separation were discussed.The results indicated that the HPCE resolution(Rs)under optimum condition was 4.26 in 8 min.Linear range of ibuprofen enantiomers’s separated front peaks is 0.053~0.942g/L,while later peak linear range is 0.178~0.942g/L.Thus ibuprofen and bupivacaine,two chiral drugs can be separated under the same condition best suited for ibuprofen.
