β-内酰胺类药物及与复方黄连注射液联合使用对金黄色葡萄球菌MSW的影响
The effect of β-lactam drugs combined with compound coptis injection to MSW of Staphylococcus aureus
【摘要】 目的测定β-内酰胺类药物单用及与复方黄连注射液联合使用对金黄色葡萄球菌ATCC29213的MIC和MPC,探讨β-内酰胺类与复方黄连注射液联合使用对耐药突变选择窗(MSW)的影响,为临床优化抗菌药物给药方案,限制耐药突变体选择性扩增提供一条新思路。方法采用MH琼脂二倍稀释法测定阿莫西林和头孢噻呋、复方黄连注射液单独用药和联合用药的最低抑菌浓度(MIC)、抑制99%接种细菌生长的最低抑菌浓度(MIC99)、防突变浓度(MPC)。结果阿莫西林单独使用时的SI(MPC/MIC99)值为15.2,与16MIC的复方黄连注射液联合使用后,SI降低至5.1,为其单独用药的1/3;头孢噻呋单独使用时其SI值为16.1,与16MIC的复方黄连注射液联合使用后,SI降低至4.2,为单独用药的1/4。结论β-内酰胺类药物和复方黄连注射液联合用药可以明显缩小MSW。
【Abstract】 Objective To determine MIC and MPC of Staphylococcus aureus 29213 to β-lactam drugs or mixture of and β-lactam drugs with compound coptis injection,investigate the effect of β-lactam combined with compound coptis injection for drug-resistant mutant selection window(MSW) and provide a new idea for optimizing regimen of antibiotics and restricting selective amplification of resistant mutants.Methods The MIC,MIC99 and MPC of amoxicillin or ceftiofur or compound coptis injection single-used as well as mixture of amoxicillin with compound coptis injection or ceftiofur with compound coptis injection were determined using method called typical MH agar double dilution.Results The SI value(MPC/ MIC99)only treated by amoxicillin was 15.2.However,if it was combined with compound coptis injection,SI value was decreased to 5.1,only 1/3 of the former.If treated with ceftiofur alone,the SI value was 16.1 when combined with compound coptis injection,the SI value was declined to 4.2,only 1/4 of the former.Conclusion Mixture of β-lactam drug with compound coptis injection can reduce MSW significantly.
