铜绿假单胞菌β-内酰胺类获得性耐药基因及外膜孔蛋白基因研究
Study on acquired β-lactamase-resistant genes and outer membrane protein gene in Pseudomonas aeruginosa
【摘要】 目的研究临床分离铜绿假单胞菌(PAE)β-内酰胺类获得性耐药基因及膜孔蛋白基因的存在状况。方法对分离自贵阳地区(A组)和江苏苏南地区(B组)PAE各20株,用纸片扩散法测定19种抗菌药物的敏感性,采用聚合酶链反应(PCR)法检测21种β-内酰胺类获得性耐药基因和膜孔蛋白基因(oprD2);对阳性基因进行测序,测序结果进行Blast分析。结果两组PAE呈β-内酰胺类、氨基糖苷类、喹诺酮类多药耐药,A组PAE检出TEM、CARB等两种β-内酰胺酶基因,阳性率均为10.0%,膜孔蛋白编码基因oprD2均为缺失,缺失率为100.0%;B组PAE检出TEM、OXA-10群、PER、GES、CARB等5种β-内酰胺酶基因,阳性率分别为55.0%、55.0%、45.0%、30.0%、10.0%,膜孔蛋白编码基因oprD2缺失率为45.0%。结论两组PAE对β-内酰胺类药物的耐药与携带β-内酰胺类获得性耐药基因和膜孔蛋白基因oprD2的缺失相关。
【Abstract】 OBJECTIVE To investigate the acquired β-lactamase-resistant genes and outer membrane protein gene in Pseudomonas aeruginosa.METHODS A total of 20 strains of P.aeruginosa isolated from Guiyang(group A) and 20 strains of P.aeruginosa isolated from Sunan area(group B) were collected,the antimicrobial susceptibility testing(AST) was tested by K-B method,and 21 acquired β-lactamase drug resistant genes and outer membrane protein gene(oprD2)were detected by polymerase chain reaction(PCR) and Blast analysis of DNA sequencing was performed.RESULTS The AST results showed that P.aeruginosa were multidrug-resistant to β-lactams,aminoglycosides and quinolones;the PCR results showed that the positive rates of TEM and CARB genes were 10.0% and the outer membrane protein gene oprD2 was not detected in group A,the positive rates of TEM,OXA-10,PER,GES and CARB genes were 55.0%,55.0%,45.0%,30.0%,and 10.0%,and the loss rate of the outer membrane protein oprD2 gene was 45.0% in group B.CONCLUSION The drug resistance of P.aeruginosa to acquired β-lactam antibiotics is related to the acquired β-lactamase genes and the loss of the outer membrane protein gene oprD2.
