2010年致白城市手足口病流行的肠道病毒71型基因特征

【摘要】 目的分析2010年致白城市手足口病流行的肠道病毒71型基因特征。方法对2010年白城市6株EV71分离株VP1编码区基因全长逆转录-聚合酶链反应扩增后进行序列测定,使用Bioedit软件和MEGA 4.0软件进行同源性、基因亲缘关系分析。结果白城市2010年的6株EV71分离株均属于C4a基因亚型,6株白城分离株间的核苷酸同源性为98.6%~100.0%,并形成2个传播链,传播链1的白城4株病毒与吉林省2010年流行株代表株JL-HFM-10-5病毒亲缘关系最近,核苷酸同源性99.7%~100.0%,传播链2的2株白城流行株与2008年的阜阳流行株和2009年的2株吉林省流行株亲缘关系最近,核苷酸同源性99.3%~99.4%。结论 2010年白城市至少有两个EV71传播链致手足口病流行,传播链1的病毒在吉林省内和白城部分县区循环传播导致手足口病的流行;传播链2的病毒已经在国内传播较长时间,提示EV71病毒跨地域迅速而广泛传播特点。

【Abstract】 Objective To analyze the genetic characteristics of enterovirus type71(EV71) isolates causing hand foot and mouth disease(HFMD) epidemics in Baicheng city in 2010.Methods 6 EV71 virus strains from the Baicheng city were chosen to do the RT-PCR and sequences of VP1 genes of EV71 strains,and analyzed by the software of bioedit and Mega 4.0.Results All 6 virus strains belonged to C4a genotype.The nucleotide homology among 6 virus strains of Baicheng city reached 98.6%-100.0%,and two transmission chain were formed.The homology which 4 Baicheng isolated strains in 2010 compared to the strain(JL-HFM-10-5) isolated in Jilin province in 2010 was the nearest which attained 99.7-100.0% among transmission chain 1.The homology which 2 Baicheng isolated strains in 2010 compared to the Fuyang strain in 2008 and the 2 strain isolated in Jilin province in 2009 was the nearest which attained 99.3%-99.4% among transmission chain 2.Conclusion At least 2 transmission chains cause HFMD epidemic in 2010 in Baicheng city,the virus strain of transmission chain 1 circulates and causes HFMD epidemic in Baicheng city and Jilin province,the virus strain of transmission chain 2 circulates longer time in China and reminds that EV71 has the characteristic with spreading rapidly and widely.
