【摘要】 目的研究VEGF基因启动子区-460T/C与-1154G/A多态性位点在中国北方汉族人群中与糖尿病足溃疡(DFU)之间的关系。方法基于病例-对照研究方法,采用PCR-RFLP技术检测209例T2DM患者(其中单纯T2DM患者112例,DFU患者97例)和健康对照(NC)者188名VEGF基因启动子-460T/C和-1154G/A的多态性,比较各组的基因型与基因频率。结果VEGF基因启动子-1154G/A多态性在患者和NC者间差异无统计学意义(P>0.05)。与单纯T2DM患者相比,DFU患者中携带C等位基因的个体频率显著降低(T/C+C/Cvs T/T,P=0.0109,OR=0.4708,95%CI=0.2625～0.8444)。DFU患者与NC者比较,携带C等位基因的个体频率也显著下降(P=0.0037,OR=0.4784,95%CI=0.2891～0.7916)。结论 DFU患者的C等位基因频率较低,可能是由于该等位基因增加了血管生成,对患者具有保护作用。更多还原

【Abstract】 Objective To investigate the association of VEGF-460T/C and-1154G/A gene polymorphism with diabetic foot ulcer in T2DM of North China Han people.Methods In this case-control study,the PCR-RFLP technique was applied to detect the VEGF-460T/C and-1154G/A gene polymorphism for comparing the genotypes and allele frequency between test and control groups.The subjects involved 209 T2DM cases(112 T2DM patients and 97 DFU patients) and 188 normal controls(NC).Results No significant difference was found in allele and genotype distributions of the-1154G/A polymorphisms between patients and controls.However,the frequency of C allele carriers at position-460 was significantly decreased in the DFU patients compared with the subjects without DFU(T/C+C/C vs T/T,P=0.0109,OR=0.4708,95%CI=0.2625~0.8444).Also there was a significant decrease in frequency of C allele carriers in DFU patients compared with the controls(P=0.0037,OR=0.4784,95%CI=0.2891~0.7916).Conclusions The decrease in frequency of C allele of the DFU patients may have a protective effect for T2DM patients,possibly because of its result in increasing angiogenesis.更多还原
