北京鸭FABP2基因多态性与体尺和屠体性状的相关性研究
Polymorphism of FABP2 Gene and Its Association with Body Size and Carcass Traits in Peking Duck
【摘要】 本研究以12个全同胞家系共224只G2代北京鸭资源群体为试验材料,分别利用DNA序列测定和PCR-RFLP方法检测G1和G2代个体基因型。结果在北京鸭小肠脂肪酸结合蛋白(FABP2)基因中共发现4处单核苷酸多态性(SNPs),外显子3中1处SNP(92bp)可导致氨基酸的改变(Gly→Ser),G2代中该SNP产生3种基因型:AA、AB和BB,其频率分别为0.103、0.629和0.268,群体中A基因频率为0.417,B基因频率为0.583。最小二乘分析表明,BB基因型个体的体斜长、胫围和颈长显著高于AB基因型;BB基因型个体胫蹼重、肌胃重和翅重显著高于AA基因型和AB基因型;BB基因型个体头重显著高于AA基因型(P<0.05)。综上所述,FABP2基因外显子3中(92bp)的SNP与北京鸭的部分体尺和屠体性状显著相关,可作为北京鸭体尺和屠体性状标记辅助选择的分子标记。
【Abstract】 The resource populations of Peking duck with a total of 224 G2 individuals containing 12 full-sib families were used in the study. DNA sequencing and PCR-RFLP were used to detect genotype G1 and G2 populations. The results showed that 4 single nucleotide polymorphisms (SNPs) were detected in intestinal fatty acid binding protein (FABP2) gene of Peking duck and one of SNPs was found at the third exon (92 bp) of FABP2 gene and the amino acid could be changed (Gly→Ser). There were three genotypes (AA, AB and BB) of the SNP in G2 populations,and their frequencies were 0.103,0.629 and 0.268,respectively,where the frequencies of the allele A was 0.417 and B was 0.583. The results of Least-square analysis showed that body length,shank girth and neck length with genotype BB were significantly higher than those with genotype AB;weight of shank and web,gizzard weight and wings weight with genotype BB were significantly higher than those with genotypes AA and AB;head weight with genotype BB was significantly higher than that with genotype AA (P<0.05). It indicated that the SNP of the third exon (92 bp) of FABP2 gene was significantly associated with some of body size and carcass traits in Peking duck and could be a molecular marker for marker assisted selection breeding on body size and carcass traits of peking duck.
