PTK787对早产儿视网膜病变大鼠模型血管内皮生长因子及其受体mRNA表达的影响
Effect of PTK787 on vascular endothelial growth factor and its receptor mRNA expressions in rat models of retinopathy of prematurity
【摘要】 目的:研究受体酪氨酸激酶亚群抑制剂PTK787对早产儿视网膜病变(ROP)大鼠模型血管内皮生长因子(VEGF)及其受体VEGFR-2 mRNA的影响。方法:建立波动氧(80%和10%氧浓度24 h交替)诱导的SD大鼠ROP模型。67只新生SD大鼠随机设立空气组、模型组、治疗组(腹腔注射PTK787 50 mg/kg);每组随机抽取6只新生鼠,利用实时定量荧光PCR,观察生后12天及第17天的视网膜组织VEGF与VEGFR-2 mRNA表达,数据采用比较CT值法(2-CT法)分析基因相对表达量。选择GAPDH为内参。结果:12天及17天模型组较对照组VEGF与VEGFR-2 mRNA表达量明显增多,而治疗组未见异常。结论:PTK787对ROP病变大鼠模型VEGF与VEGFR-2 mRNA表达呈明显的负性调控作用。
【Abstract】 Objective:To study the effect of PTK787 on vascular endothelial growth factor(VEGF) and its receptor-2(VEGFR-2) mRNA expressions in rat models of retinopathy of prematurity(ROP).Methods:SD rat models of ROP were established by induction of alternative oxygen concentrations(80% oxygen and 10% oxygen alternation every 24 hours).A total of 67 neonatal SD rats were divided into air group,model group,and treatment group(intraperitoneal injection of PTK787 50 mg/kg);six neonatal rats were selected from each group randomly,quantitative fluorescent PCR was used to observe the expressions of VEGF mRNA and VEGFR-2 mRNA in retinal tissues at 12 and 17 days after birth,the data were compared by 2-CT method,and the relative expression value of gene was analyzed.GAPDH mRNA was used as the internal control.Results:The expression levels of VEGF mRNA and VEGFR-2 mRNA at 12 and 17 days after birth in model group were significantly higher than those in control group,while in treatment group,there was no abnormality.Conclusion:PTK787 plays a significant down-regulating role for expressions VEGF mRNA and VEGFR2 mRNA in rat models of ROP.
