浙贝乙素胆酸盐中浙贝乙素和胆酸的含量测定

【摘要】 目的建立浙贝乙素胆酸盐中浙贝乙素和胆酸的含量测定方法。方法以甲醇-乙酸铵(0.1 mol.L-1)-三乙胺-冰乙酸(78∶22∶0.2∶0.03)为流动相,C18柱(250 mm×4.6 mm,5μm)分离,流速为1 mL.min-1;蒸发光散射检测器检测(漂移管温度72.1℃,氮气流速2 mL.min-1)。结果浙贝乙素胆酸盐中浙贝乙素和胆酸分别在50.4~151.1μg.mL-1(r=0.999 6,n=5)、50.5~151.4μg.mL-1(r=0.999 9,n=5)内峰面积与浓度呈良好线性关系;平均回收率分别为99.17%(RSD=0.62%)和99.37%(RSD=0.63%)。结论该方法测定浙贝乙素胆酸盐中的浙贝乙素和胆酸含量简便,可靠,重复性好。

【Abstract】 Objective To establish an HPLC method for determination of verticinone and cholic acid in verticinone-cholic acid salt.Methods The separation was performed on a C18 column(250 mm×4.6 mm,5 μm).The mobile phase was methanol : ammonium acetate(0.1 mol·L-1) : triethylamine : ethanoic acid(78:22:0.2:0.03).The flow rate was 1 mL·min-1.The gas flow rate of ELSD was 2 mL·min-1 and the temperature of drift tube was 72.1 ℃.Results The linear ranges of verticinone and cholic acid were 50.4-151.1 μg·mL-1(r=0.999 6,n=5) and 50.5-151.4 μg·mL-1(r=0.999 9,n=5) respectively.The average recoveries of the two components were 99.37%(RSD=0.63%,n=9) and 99.17%(RSD=0.62%,n=9) respectively.Conclusion The method is simple and reliable for the quality control of verticinone and cholicacid salt.
