复方松及片提取物对博莱霉素诱导的小鼠肺纤维化的影响
Influence of the extract of Compound Formula Rosin and Bletilla Table in Bleomycin induced pulmonary fibrosis in mice
【摘要】 目的观察复方松及片(CFRB)对博莱霉素诱导的小鼠肺纤维化的影响,初步探究复方松及片对肺纤维化的作用及其机制。方法采用气道滴入博莱霉素(2.5 mg/kg)制备肺纤维化模型,在造模后每日采用灌胃方式给予复方松及片提取物,对照组腹腔注射地塞米松。连续用药14 d后,采用生理盐水进行支气管肺泡灌洗(bronchoalveolarlavage fluid,BALF),收集BALF,ELISA法测定转化生长因子-β1(transforming growth factor-β1,TGF-β1)的含量。液氮速冻肺组织,-80℃保存,检测各组肺组织羟脯氨酸含量,以及PDE8A mRNA的表达。结果复方松及片提取物对博莱霉素气道滴入诱导的肺纤维化变化有一定的抑制作用,能改善肺组织的纤维化病理改变。复方松及片提取物能明显降低BALF中TGF-β的含量,且对磷酸二酯酶8A(PDE8A)的mRNA表达也有一定的抑制作用。结论复方松及片对博莱霉素诱导的肺纤维化有一定作用,提示复方松及片可能用于肺纤维化的治疗,同时PDE8A可能是肺纤维化的新靶点。
【Abstract】 Objective To observe the effect of the extract of Compound Formula Rosin and Bletilla Table(CFRB) on Bleomycin-induced pulmonary fibrosis in mice,initially explore the mechanisms of the extract.Methods Belomycin(2.5 mg/kg) induced pulmonary fibrosis in mice were used,CFRB extracts were administrated by daily gavage,control group was intraperitoneal injection of dexamethasone.After continuous treatment for 14 d,bronchoalveolar lavage and lung tissue specimens were collected.Transforming growth factor-β1(TGF-β1) levels in BALF were determined by ELISA;hydroxyproline contents were determined by assay kit;PDE8A mRNA expression in mouse lung tissue was detected by real time-PCR.Results CFRB extract had a certain inhibitory effect on the changes of the airway of Bleomycin instillation induced pulmonary fibrosis,and could improve the lung tissue of fibrosis change.CFRB extract could significantly reduce the content of the BALF of TGF-β1,and might inhibit the mRNA expression of PDE8A.Conclusion CFRB has anti-fibrosis action in Belomycin induced mice model,which suggesting that the compound may be used for the treatment of pulmonary fibrosis,at the same time PDE8A may be a new target for pulmonary fibrosis.
