Y染色体微缺失与精子发生的相关性分析
Microdeletions of AZF in Y chromosome and spermatogenesis failure in infertile men
【摘要】 目的:探讨Y染色体AZF微缺失与男性精子发生之间的关系。方法:采用聚合酶链反应技术对343例无精子症(包括精子发生基本正常177例和生精障碍166例)、633例严重少精症、45例严重弱精症患者进行AZFa、AZFb、AZFc等3个区域微缺失分析。结果:166例生精障碍患者中发现AZF微缺失41例,总缺失率为24.7%;177例生精基本正常的男性发现3例合并AZFc的微缺失,缺失率为1.7%;在严重少精组发现AZF微缺失49例,总缺失率为7.7%;弱精症患者无一例发现AZF微缺失。生精障碍组与其他男性比较Y染色体AZF区域微缺失率差异有统计学意义(P<0.01)。结论:Y染色体AZF区域微缺失与男性生精障碍有着明显的相关性,因此有必要对门诊的生精障碍患者以及准备行辅助生育技术的生精障碍患者行Y染色体AZF微缺失筛查。对于这些患者是否需要进行PGD技术还需要更多的数据证明。
【Abstract】 Objectives: To explore the relationship between AZF microdeletions in Y chromosomal and male spermatogenesis.Methods: The study recruited 1021 infertile Chinese men,including 343 men of azoospermia(177 with nearly normal spermatogenesis in tests and 166 cases with maturation arrest in tests),633 men of oligozoospermia and 45 men of asthenospermia.Y chromosomal microdeletions in AZFa,b,c regions were screened with a multiplex polymerase chain reaction(PCR).Results: In azoospermia patients,41(24.7%) cases with Y chromosomal microdeletions were found with spermatogenesis failure,while 3 cases were found to have normal spermatogenesis.There were 49 cases with AZF microdeletions found in oligozoospermics.No case was found in asthenospermics.There was a significant difference in Y chromosomal microdeletions between the patients with spermatogenesis failure and other infertile men(P<0.01).Conclusion: Microdeletion of AZF was a genetic risk factor for spermatogenic failure and male infertility.Therefore,patients of zoospermia and oligozoospermia.should be screened for Y chromosome microdeletion.
