南瓜叶中γ-氨基丁酸的提取及薄层扫描测定
Determination and Extraction of γ-Aminobutyric Acid from Pumpkin(Cucurbita mosehata) Leaves
【摘要】 本文在单因素试验基础上,通过正交试验优化了南瓜叶中γ-氨基丁酸的提取条件,并对γ-氨基丁酸的薄层扫描测定方法进行了探索。结果表明,南瓜叶中γ-氨基丁酸的最佳提取条件为:以20%乙醇为溶剂、料液比1∶17(w/v)、提取时间1 h,此条件下得到的γ-氨基丁酸的含量为209 mg/100 g。所建立的薄层扫描测定方法在0.125~2.0 mg/mL范围内呈良好线性关系(R2=0.9991),平均加样回收率(n=4)为99.61%。
【Abstract】 The extraction conditions and thin layer chromatography scanning(TLCS) analytic method of γ-aminobutyric acid from pumpkin leaves were studied.On the basis of single factor experiments,orthogonal test was investigated.The optimum parameters for extracting γ-aminobutyric acid from pumpkin leaves were determined as follows:the concentration of alcohol was 20%,the ratio of solid to liquid was 1∶ 17(w/v),and the extraction time was 1 h.Under these conditions,the yield of γ-aminobutyric acid was 209 mg/100 g.The calibration curve showed good linearity in the range of 0.125 to 2.0 mg/mL(R2 =0.9991) with the average recovery(n=4) of 99.61%.
