利用P5CS基因转化白三叶的研究
Transformation of White Clover(Trifolium repens L.) by P5CS Gene
【摘要】 为了培育抗旱白三叶,构建了植物表达载体pBPC-P5CS,利用基因枪法转化白三叶的愈伤组织,PCR检测和Southern blot鉴定证实白三叶中已导入P5CS基因。对转P5CS基因白三叶植株的不同抗旱指标进行了分析。结果表明,与对照相比,转P5CS基因株系的抗旱能力得到了较大的提高。干旱胁迫下,与对照相比,转P5CS基因植株的脯氨酸含量和相对含水量分别比对照高20.0%-21.2%和5.6%-8.5%。
【Abstract】 In order to cultivate drought resistance white clover(Trifolium repens L.),the plant expression vector pBPC-P5CS was cons-tructed and transformed into calli of white clover via microprojectile bombardment method.The PCR amplification.Southern blot method were used to identified the P5CS gene in the transgenic white clover.The different drought resistance index of transgenic P5CS white clover plants was analyzed.The results showed that the transgenic plants can enhance the drought resistance comparing to control plants.Under drought stress,the proline content and RWC of transgenic plants was 20.0%-21.2%,5.6%-8.5% higher than that that of wild type(WT),respectively.
