23株芽孢杆菌16S rRNA基因序列扩增与系统发育分析
Identification of 23 Bacillus Strains by Phylogenetic Analysis of 16S rRNA Gene Sequence
【摘要】 采用16S rRNA基因序列分析对西南菌种站(SICC)收集保藏的23株芽孢杆菌进行复核鉴定,以改良的CTAB法提取细菌DNA,用通用引物F1、R1对菌株进行16S rRNA扩增,使用Clustal X对序列进行比对分析,并用MEGA 4进行建树分析。鉴定结果表明其中13株与原学名一致,1株与原学名不符,其余9株在绘制的N-J发育树中显示同时与枯草芽孢杆菌(Bacillus subtilis)、解淀粉芽孢杆菌(B.amyloliquefaciens)汇聚到一个分支,需要用特异PCR方法进一步核实。
【Abstract】 We identified 23 Bacillus strains from SICC by 16S rRNA gene sequence analysis.The genomic DNA was isolated and purified with the improved CTAB methods.The sequences of 16S rRNA were amplified by PCR with the bacterium universal primers and the purified PCR products were directly sequenced.The correct sequences were aligned with Clustal X and the phylegenetic tree was constructed by MEGA 4.13 strains support the original identification results,one strains belong to Bacillus circulans,B.subtilis and B.amyloliquefaciens need to identify with specific PCR.
