基质分散固相萃取净化液相色谱检测谷物中赭曲霉毒素A

【摘要】 建立了经基质分散固相萃取(MSPD)净化,使用液相色谱仪配荧光检测器分析谷物中赭曲霉毒素A含量的方法。C18固定相与粉碎后的样品置于研钵中,使用研棒轻柔的充分混合,将研磨后的混合物置于带有筛板的玻璃注射器管中,用10ml甲醇淋洗、定容,收集淋洗液经过0.45μm滤膜过滤。滤液直接使用配有荧光检测器(FLD)液相色谱仪分析。结果显示,在选定色谱条件下,方法对谷物中含有的赭曲霉毒素A可以有效分离。回收率为80.0%~93.65%,最低检测限为0.5μg/kg。表明该方法能够快速、准确地检测谷物中赭曲霉毒素A的含量。

【Abstract】 To establish a method of extraction with matrix solid-phase dispersion(MSPD),high performance liquid chromatography(HPLC) coupled with fluorescence detection for determination of ochratoxin A(OTA) in cereal.C18 solid-phase and samples powder placed in the mortar gently and fully mixed by pestle.And then put in glass syringes with sieve plate,rinsed by 10 ml methanol,and collect the eluent through 0.45 μm membrane filter.The filtrate analysis by high performance liquid chromatography coupled with fluorescence detection.The results showed that ochratoxin A in cereal can be an effective separation in selected conditions,recovery rate is between 80.0% to 93.65%,the limit of detection is 0.5 μg/kg.These indicated that this method could detect the additive contents of ochratoxin A in cereal,accurately and simultaneously.
