SDF-1/CXCR4生物轴在喉鳞状细胞癌中的表达和意义

【摘要】 目的:观察喉鳞状细胞癌(LSCC)中基质细胞衍生因子(SDF-1)及其受体融合素即CXCR4的表达情况,研究LSCC中SDF-1与CXCR4表达的相关性,并探讨其在LSCC发生、发展及淋巴结转移过程中的作用和可能机制。方法:应用免疫组织化学法(SP法)检测SDF-1及CXCR4在LSCC组织、癌旁组织、正常喉黏膜及颈部淋巴结中的表达情况,运用Kruskal-Wallis检验、χ2检验、Spearman等级相关等方法进行统计分析。结果:SDF-1及CXCR4在LSCC中的阳性表达率均显著高于癌旁组与正常组(P<0.01);SDF-1及CXCR4的表达与淋巴结转移(P<0.01)、临床分期(P<0.01)和病理分级(P<0.05)有相关性;SDF-1及CXCR4在阳性淋巴结组织中的表达率均高于阴性淋巴结组织(P<0.01);LSCC中SDF-1与CXCR4的表达呈正相关。结论:SDF-1及CX-CR4蛋白在喉癌组织及阳性淋巴结组织中高表达,并与淋巴结转移、临床分期、病理分级有相关性,提示二者可能与LSCC的浸润及转移有关,并且在LSCC的发生、发展中有协同作用。

【Abstract】 Objective:To study the correlation between the stromal cell-derived factor(SDF-1)and the receptor fusin(CXCR4)in carcinoma of larynx,and investigate some mechanisms of SDF-1/CXCR4during the development,invasion and lymph node metastasis of laryngocarcinoma.Method:Detecting the expression of SDF-1and CXCR4by immunohistochemical method(SP)in laryngocarcinoma,paraneoplastic tissues,normal laryngeal mucosa and cervical lymph node.Using Kruskal-Wallis H test,χ2 test,Spearman rank correlation analysis and so on to do statistical analysis.Result:The positive expression rate of SDF-1and CXCR4in laryngocarcinoma was obviously higher than in paraneoplastic tissues and normal laryngeal mucosa tissues(P<0.01).And the expression of two proteins was correlated with lymph node metastasis(P<0.01),clinical stage(P<0.01)and pathological grading of tumor(P<0.05).The positive expression rate of SDF-1and CXCR4protein in metastasis lymph node tissue was higher than that in non metastasis lymph node tissue(P<0.01).The expression of SDF-1is correlated positively with the expression of CXCR4in laryngocarcinoma.Conclusion:SDF-1and CXCR4protein are highly expressed in laryngocarcinoma and in metastasis lymph node tissue.And they are correlated with lymph node metastasis,clinical stage and pathological grading of the tumor.According to the results,the two proteins may relate to infiltration and metastasis of laryngeal squamous cell carcinoma and play a role of synergistic action in the development and invasion of carcinoma of larynx.
