不同组织来源单核/巨噬细胞对伴放线放线杆菌吞噬能力的比较研究

【摘要】 目的:比较血液单核细胞(monocytes,Mo)、肺泡巨噬细胞(alveolar macrophage,AM)、腹腔巨噬细胞(peritoneal macrophage,PM)和肝脏枯否细胞(kuffer cells,KC)对伴放线放线杆菌(Actinobacillus actinomycetemcomitans,Aa)吞噬能力的差别。方法:3只健康新西兰兔,分离培养Mo、AM、PM和KC;荧光素标记Aa,以感染复数25∶1的比例感染细胞;流式细胞术检测0.5、1.0、1.5h时吞噬细菌的阳性细胞率和平均荧光强度,计算吞噬指数。结果:1.5h内,随着孵育时间增加,AM和PM对Aa吞噬能力逐渐升高,Mo和KC的吞噬能力在1h达到高峰;Mo、AM、PM和KC吞噬功能存在明显差异,孵育0.5h吞噬指数AM、PM>Mo、KC,1.0h时AM>PM>Mo、KC,1.5h时AM>PM>KC>Mo。结论:不同部位单核/巨噬细胞对Aa的吞噬能力不同,可能是造成牙周病对不同部位影响不同的重要原因。

【Abstract】 Objective:To compare the phagocytosis ability of Actinobacillus actinomycetemcomitans(Aa) by peripheral mononuclear cells(Mo),alveolar macrophage(AM),peritoneal macrophage (PM) and kuffer cells (KC). Methods:Mo,AM,PM and KC from 3 healthy New Zealand rabbits were isolated and purificated. Aa was lableded with fluorescein isothiocyanate (FITC). Monocytes/macrophages were infected with FITC-Aa in the ratio of 25∶1(25 bacteria∶1 cell). The positive cell rates and mean fluorescent intensity (MFI) were detected by flow cytometry at 0.5,1.0,1.5 h. Phagocytic index was calculated. Results:Within 1.5 h,the phagocytic indexes of AM,PM rose as the time increased; the phagocytosis ability of Mo and KC reached the peak at 1 h. Obvious difference of phagocytosis among Mo,AM,PM and KC was observed. At 0.5 h,the phagocytic indexes were : AM and PM>Mo,KC; at 1.0 h,AM>PM>Mo and KC; at 1.5 h,AM>PM>KC>Mo. Conclusions:The different phagocytosis of Aa by monocytes/macrophages of various tissues may be a important reason why periodontitis has different effects on different parts.
