丹栀逍遥散防治多囊卵巢综合征模型大鼠的P450arom及ERK通路研究
Effects of Dan Zhi Xiao-Yao Power on rat polycystic ovary syndrome and its regulation of P450arom and ERK 1/2
【摘要】 目的:观察丹栀逍遥散(DZXYP)对多囊卵巢综合征(PCOS)模型大鼠的作用及机制。方法:用皮下埋植左旋18-甲基炔诺酮硅胶棒联合皮下注射人绒毛膜促性腺激素的方法诱导PCOS模型,化学发光法检测血清黄体酮(LH)、卵泡刺激素(FSH)、雌二醇(E2)和睾酮(T),免疫组化检测细胞色素P450芳香化酶(P450arom)的表达,western blotting检测P450arom及细胞外信号调节激酶(ERK)1/2的活化。结果:DZXYP降低LH(P<0.05)和T(P<0.01)水平,增高FSH水平(P<0.01);增加P450arom的表达,降低ERK 1/2磷酸化。结论:P450arom表达降低是大鼠该PCOS模型发病的因素之一,DZXYP可能通过抑制ERK 1/2磷酸化,增加P450arom表达防治PCOS。
【Abstract】 Aim:To investigate the effects of DanZhi XiaoYao Power(DZXYP) on rat polycystic ovary syndrome(PCOS) rats and its regulation on aromatase cytochrome P450(P450arom) and extracellular signal-regulated kinase ERK 1/2.Methods: Capsulae Levonorgestreli Silasticus and human chorionic gonadotrophin were used to induce PCOS model in rats.LH,FSH,E2 and T were measured by chemiluminescence.Immunohistochemistry was used to analyze the expression of P450arom and Western blotting was used to determine the expresson of P450arom and phosphorylation of ERK 1/2.Results: DZXYP reduced the level of LH,T and increased the level of FSH.It also increased the expression of P450arom and decreased the phosphorylation of ERK 1/2.Conclusion:The lowered expression of P450arom is an important factor in the pathogenesis of the PCOS model.DZXYP might ameliorate PCOS by increasing P450arom expression and reducing ERK 1/2 phosphorylation.
