贵州马铃薯栽培品种遗传多样性的SRAP分析
Genetic Diversity Analysis of Potato Cultivars in Guizhou by SRAP Marker
【摘要】 为辅助育种中合理选配亲本,减少育种的盲目性,加快遗传改良进程提供可靠依据,采用相关序列扩增多态性(SRAP)分子标记对11份马铃薯栽培品种的遗传多样性进行分析,利用随机的40对SRAP引物对11份马铃薯品种的基因组DNA进行特异扩增。结果表明:20对引物扩增出55个多态性条带,多态性引物比达24%,平均每对引物产生2.9个多态性条带;11份马铃薯品种间的SRAP标记遗传相似系数为0.18~0.95,当遗传相似系数为0.53时,将11份品种分为2大类群,其中,第Ⅰ类群可进一步分为Ⅰ-A和Ⅰ-B 2个亚簇;聚类分析表明,参试马铃薯品种间的遗传多样性相对丰富,遗传差异较大。
【Abstract】 To select parents rationally,reduce the blindness of breeding,so as to provide reliable references for accelerating the genetic improvement process,the sequence related amplified polymorphism(SRAP) was used to analyze the genetic diversity of 11 potato cultivars.Meanwhile,the specific amplification of genomic DNA from 11 potato cultivars was conducted using randomly selected 40 primer pairs.The results showed that twenty primer pairs were polymorphic,and the polymorphism rate of primers was 24%.55 bands were obtained,2.9 polymorphism bands per primer pairs on average.The genetic distance of 11 potato varieties ranged from 0.18 to 0.95.At the genetic coefficient of 0.53,11 potato varieties were divided into two groups.Furthermore,the group Ⅰ could be subdivided into subgroupsⅠ-A and Ⅰ-B.The result of cluster analysis showed that the genetic diversity of the tested potato cultivars varied widely.And the genetic difference was wide.
