N-甲基-N-顺式-苯乙烯基-桂皮酰胺对青霉素诱发惊厥大鼠神经细胞内钙离子含量的影响
Effects of N-methyl-N-cis-styryl-cinnamamide on intracellular calcium ion concentration(cell[Ca2-] i) in seizure rats induced by penicillin
【摘要】 目的:检测黄皮核提取物N-甲基-N-顺式-苯乙烯基-桂皮酰胺(N-methyl-N-cis-styryl-cinnamamide,s-MSC)对皮层定位注射青霉素(Penicillin,PNC)诱发惊厥大鼠脑神经细胞内钙离子含量的影响,初步探讨其抗惊机理。方法:20只大鼠分为5组:正常对照组(20 ml/L吐温80+生理盐水)、模型对照组(20 ml/L吐温80+PNC)、丙戊酸钠120 mg/kg+PNC7、5 mg/kg s-MSC组(s-MSC 75 mg/kg+PNC)、150 mg/kg s-MSC组(s-MSC 150 mg/kg+PNC)。建立皮层定位注射PNC诱发大鼠癫痫模型,流式细胞术检测各组大鼠左侧大脑初级、次级躯体运动皮层及左侧海马神经细胞内钙离子含量。结果:皮层定位注射PNC后大鼠脑神经元内钙离子含量增加(P<0.01),丙戊酸钠及150 mg/kg的s-MSC均可使致癫大鼠脑神经元内钙离子含量降低(P<0.01)7,5 mg/kg s-MSC对于细胞内钙离子的降低无统计学意义。结论:黄皮核提取物N-甲基-N-顺式-苯乙烯基-桂皮酰胺对抗大鼠皮层定位注射PNC诱发的惊厥可能与其降低神经元内钙离子含量有关。
【Abstract】 To investigate the effect of the seed extract of Clausena lansium,N-methyl-N-cis-styryl-cinnamamide,on intracellular calcium ion concentration(cell[Ca2-] i) in seizure rats induced by penicillin and explore its mechanism of anticonvulsant action.Method:Twenty Sprague-Dawely rats were randomly divided into five groups with 4 rats in each group:normal control(20 ml/L Tween 80 + Normal Saline),model control(20 ml/L Tween 80 + Penicillin),positive control(Sodium Valproate 120 mg/kg + Penicillin),low dose treatment(s-MSC 75 mg/kg + Penicillin),high dose treatment(s-MSC 150 mg/kg + Penicillin).The seizure rat model was established by penicillin localized injected in cortex.The change of intracellular calcium ion concentration(cell[Ca2-] i) in rats neuron of the left side of the primary,secondary somatic motor cortex and left hippocampus was detected by flow cytometry.Result:The neuronal intracellular calcium ion concentration(cell[Ca2-] i) of rat cerebral cortex was significantly increased after penicillin injection,compared with the normal control group(p<0.01).Compared with the model group,high dose N-methyl-N-cis-styryl-cinnamamide and Sodium Valproate can significantly decreased intracellular calcium ion concentration(cell[Ca2-] i)(P<0.01).However,there was no significant difference in intracellular calcium concentration(cell[Ca2-] i) between the model and low dose cinnamamide treatment groups.Conclusion:The anticonvulsive function of the seed extract of Clausena lansium,N-methyl-N-cis-styryl-cinnamamide,might relate with decreasing intracellular calcium ion concentration(cell[Ca2-] i) level.
