中药补肾宁对肾阳虚模型大鼠卵巢MMP-9mRNA的影响

【摘要】 目的:探讨"补肾宁"对氢化可的松法复制肾阳虚模型大鼠卵巢MMP-9mRNA的影响。方法:成年雌性SD大鼠60只,随机分为6组。分别为空白对照组、模型对照组、克罗米芬组、补肾宁低、中、高剂量组。空白对照组每日肌注等容积生理盐水,其余5组,每日下肢肌注氢化可的松注射液,连续11天,制备肾阳虚模型大鼠。11天后按组别分别用药治疗,治疗期为15天。采用原位杂交技术观察各组卵巢局部MMP-9mRNA表达的变化。结果:模型组卵巢MMP-9mRNA的阳性表达明显低于其它各组,其中与补肾宁中、高剂量组比较差异显著(P<0.01);补肾宁中、高剂量组与空白对照组比较MMP-9mRNA表达强度无显著性差异(P>0.05);补肾宁高剂量组与克罗米芬组、补肾宁低剂量组比较有显著性差异(P<0.05)。结论:肾阳虚模型大鼠卵巢MMP-9mRNA表达减弱,可能与卵泡发育异常及排卵障碍有关。补肾宁能够增强肾阳虚模型大鼠卵巢MMP-9mRNA的表达,起到促卵泡发育和促排卵的作用。

【Abstract】 Objective:To observe the effect and mechanism which BuShenNing(BSN)decoction promoted the ovulation of kidney-yang defieiency female rat model,which was replicated by applying hydoreortisone.Method:The sixty mature female Sprague-Dawley rats were divided into six groups randomly:normal contrast group,model contrast group,clomifene group,BSN low,middle and high dosage group.Normal contrast group used physiological saline,the other groups replicated kidney-yang defieiency model by muscle injecting hydoreortisone 36mg/kg,for 11 day.After 11 days,each grop used the corresponding medicine treatment for 15 days.The expression of ovarian matrix metalloproteinase-9 Mrna(MMP-9mRNA) was observed.Results: Model contrast group significantly lower than the other groups in the expression of ovarian MMP-9mRNA,BSN middle and high dosage group were significantly superior to model contrast group(P<0.01),were similar to normal contrast group(P>0.05),and BSN high dosage group was superior to clomifene group and BSN low dosage group(P<0.05).Conclusions: The expression of ovarian MMP-9mRNA was weakened in yang deficiency rat model,which may be related to abnormal follicular development and ovulation disorders.BSN groups could enhance the expression of ovarian MMP-9mRNA and promote follicular stimulating ovarian development.
