【摘要】 目的建立测定大鼠血浆中槲皮素含量的高效液相色谱方法(high performance liquid chyomatog-raphy,HPLC)。方法采用依利特Hypersil BDS C18柱,SD大鼠血浆,以甲醇:乙腈:磷酸:水(20:25:5:50)为流动相,检测波长:255nm。结果在0.1～22.0μg/mL范围内,槲皮素的峰面积与浓度呈良好线性关系,r=0.997;方法平均回收率为96.4%～103.7%;方法重现性较好,日内RSD≤7.3,日间RSD≤7.5。结论该法简单,快速,稳定,灵敏度高,重现性好,可用于槲皮素在大鼠血浆中的含量测定。更多还原

【Abstract】 【Objective】 To construct a HPLC method for the determination of quercetin in the rat’s serum.【Method】 The assay was conducted on a Hypersil BDS C18 column with methanol-20%: acetonitrile-25%: phosphoric acid-5%: water-50% as mobile phase and SD rat’s serum as a sample.The detection wavelength was 255 nm.【Result】 The linear concentration range of quercetin was 0.1~22 μg/mL,with the recovery of 96.4%～103.7%.This method has a good reproducibility.Within-day RSD≤7.3 and Between-day RSD≤7.5.【Conclusion】 The method which has a high sensitivity and a good reproducibility is simple and stable to carry out.The method may be used for the determination of quercetin in the rat’s serum.更多还原
