辐照-纤维素酶糖化降解芒草工艺条件优化研究

【摘要】 以芒草(Miscanthus.sinensis Andress)为试验材料,研究60Co-γ辐照和纤维素酶对其糖化效率的影响。通过对辐照剂量、加酶量、酶解温度、酶解时间和固液比5个因素进行单因素试验,对芒草酶解液中还原糖含量进行检测,最后通过正交试验对酶解条件进行优化。试验结果表明:未经辐照时,当加酶量为240U/g,酶解温度40℃,酶解时间60h,固液比1∶65,芒草的还原糖产量仅为132.59mg/g;而当辐照剂量为800kGy,加酶量150U/g,酶解温度40℃,酶解时间60h,固液比1∶55,还原糖产量可达281.37mg/g。

【Abstract】 The effects of 60Co-γ irradiation and cellulose hydrolysis on the saccharification of Chinese silvergrass were investigated in this paper.The optimal conditions including irradiation dosage,enzyme amount,enzyme hydrolysis temperature,enzyme hydrolysis time and solid/liquid ratio were obtained by orthogonal experiments.The results showed that the reducing sugar production was 132.59mg/g when Chinese silvergrass was treated with 270U/g cellulase at 40℃ for 60h and solid/liquid ratio was 1∶65 without irradiation.The reducing sugar production was 281.37mg/g when Chinese silvergrass was treated with 800kGy irradiation and 150U/g cellulase at 40℃ for 60h and solid-liquid ratio was 1∶55.
