【摘要】 目的筛选和确定新的膀胱癌标志物,探索更好的非侵入性的诊断膀胱癌的方法。方法通过生物信息学筛选,获得候选膀胱癌标志物基因,然后通过RT-PCR和Western Blot进行验证,确定其表达特性,并通过DotBlot初步确定其作为膀胱癌新标志物的临床价值。结果通过生物信息学对人类基因组超过50000个基因进行筛选,获得候选的膀胱癌标志物基因,然后进行RT-PCR试验验证,结果发现PKS13的mRNA在膀胱癌组织表达明显高于癌旁正常组织,且在全身其他正常组织中基本没有表达,呈现明显的膀胱癌特异高表达的特性;通过WesternBlot试验验证,结果发现PKS13的蛋白在膀胱癌组织表达明显高于癌旁正常组织,呈现明显的膀胱癌高表达特性。进一步通过DotBlot检测发现:膀胱癌患者尿液PKS13蛋白水平明显高于正常对照组。结论 PKS13是一个较好的膀胱癌标志物,可能在膀胱癌早期诊断、术后监测中具有重要的临床应用价值。更多还原

【Abstract】 Objective To screen and assess new bladder cancer marker and explore a better non-invasive method for diagnosis of bladder cancer.Methods Candidate genes were obtained with bioinformatics screening.RT-PCR and Western Blot were used here to identify the expression profile of the selected candidate genes.The clinical value of candidate markers was assessed with Dot Blot.Results By bioinformatics screening in more than 50 000 genes of human genome,we obtained several candidate genes.We chose genes to test with RT-PCR and Western Blot.RT-PCR and Western Blot analysis showed that PKS13 highly expressed in tissues of bladder cancer than adjacent normal tissues and scarcely expressed in other 16 normal tissues.Furthermore,the urinary PKS13 levels are uniformly elevated in bladder cancer patients than those of normal volunteers.Conclusions PKS13 is a new bladder cancer marker.The expression profile of PKS13 indicated that the important value of PKS13 in the early detection and monitoring of bladder cancer.更多还原
