组织工程化骨髓基质细胞与新型多孔支架材料复合体的体外研究

【摘要】 目的探讨新型多孔支架材料3-羟基丁酸-co-3-羟基戊酸共聚物/溶胶-凝胶生物活性玻璃(PHBV/SGBG)对狗骨髓基质细胞(BMSCs)生物学功能的影响。方法采用体外培养技术,将狗BMSCs与PHBV/SGBG支架材料在体外复合培养,通过扫描电镜观察狗BMSCs在PHBV/SGBG材料中的生长情况,用MTT法、碱性磷酸酶活性检测法检测PHBV/SGBG对狗BMSCs增殖、分化的影响。结果狗BMSCs可紧密附着于材料表面,黏附生长、增殖,并有突起向材料的连通微孔内伸展。复合培养3、6d,PHBV/SGBG对狗BMSCs的增殖、分化表现出一定的促进作用。结论 BMSCs与PHBV/SGBG支架材料复合体具有良好的生物学功能,有望应用于牙周组织工程学。

【Abstract】 Objective To investigate the biological effects of the cultured dog bone marrow stromal cells(BMSCs) with the novel porous scaffold poly(3-hydroxybutyrate-co-3-hydroxyvalerate)/sol gel bioactive glass(PHBV/SGBG).Methods Dog BMSCs were cultured on the PHBV/SGBG scaffolds in vitro.The cell attachment and distribution on the scaffold were observed under a scanning electron microscope(SEM).MTT assay and alkaline phosphatase(ALP) activity measurement were used to detect the effects of the PHBV/SGBG scaffolds on cell proliferation and differentiation.Results Dog BMSCs firmly attached to and grew well on the surface of PHBV/SGBG scaffolds.Obvious cell processes stretched into the micro-pores structure.After 3-day-and 6-day-culture with the PHBV/SGBG scaffolds,the proliferation and differentiation of dog BMSCs were improved.Conclusion It suggested that the compound of dog BMSCs and PHBV/SGBG scaffold with good biological properties would be a potential method for periodontal tissue engineering.
