单侧甲状腺电凝毁损法构建大鼠亚临床甲状腺功能减退症模型

【摘要】 目的评价单侧甲状腺电凝毁损法建立大鼠亚临床甲状腺功能减退症模型的可行性。方法 36只Wistar大鼠随机分为假手术组、亚临床甲状腺功能减退症组和临床甲状腺功能减退症组。亚临床甲状腺功能减退症组大鼠电凝毁损单侧甲状腺,临床甲状腺功能减退症组大鼠电凝毁损全部甲状腺,假手术组暴露甲状腺而不毁损。饲养2周后,相同时间组间平行采血、化学发光法检测血清TSH、fT4、T3水平,光镜下观察亚临床甲状腺功能减退症组大鼠对侧甲状腺组织病理形态学变化。结果与假手术组比较,大鼠单侧电凝毁损甲状腺术后2周血清TSH水平轻度升高而T3、fT4则无显著变化,残留甲状腺组织轻度增生,符合亚临床甲状腺功能减退症的特征;大鼠双侧电凝毁损甲状腺2周后血清T3、fT4显著下降、TSH水平显著升高,符合临床甲状腺功能减退症的诊断标准。结论单侧甲状腺电凝毁损法建立大鼠亚临床甲状腺功能减退症模型是用于研究亚临床甲状腺功能减退症发生及发展机制的简便可靠模型。

【Abstract】 Objective To investigate the feasibility of hemi-thyroid electrocoagulation in the establishment of a rat model of subclinical hypothyroidism(SCH).Methods Thirty-six young Wistar rats were randomly divided into three groups including sham,subclinical hypothyroidism,and clinical hypothyroidism(CH) groups.The rats in SCH group were given hemi-thyroid electriocoagulation,rats in CH group were given total thyroid electrocoagulation,while rats in the sham group only exposed thyroid without electrocoagulation.2 weeks after operation,blood samples of all rats were collected and serum thyrotropic-stimulating hormone(TSH),free thyroxine(fT4),and triiodothyronine(T3) were detected by chemolu-minescence,while the thyroid tissue specimens were examined by histopathology using hematoxylin and eosin(HE) stai-ning.Results Compared with the sham group,the weight of rats in CH group decreased,while there was no significant difference between the SCH group and sham group.The TSH level in both SCH and CH groups was significantly increased,the serum fT4 and thiiodothyronine T3 level of CH rats decreased significantly,while there was no significant difference be-tween the SCH group and sham group.The pathology revealed slight hyperplasia in the contralateral thyroid of SCH rats.Conclusions The establishment of SCH rat model by hemi-thyroid electrocoagulation is simple and convenient for longitu-dinal study on the mechanism of initiation and progress of subclinical hypothyroidism.
