两种实验优化方法的比较研究:在制备薄荷油-β-环糊精包合物过程中的应用

【摘要】 目的比较正交实验与box-behnken实验两种实验设计方法在制备薄荷油-β-环糊精包合物过程中的应用,以期对两种实验设计方法进行合理选择。方法采用饱和水溶液法制备包合物,考察因素为β-环糊精/薄荷油比例(w/v)(X1)、包合时间(X2)与包合温度(X3),以包封率和收率为考察指标,在薄荷油-β-环糊精包合物实验设计中应用上述两种设计方法,优化制备薄荷油包合物最佳条件,并对两种方法得到的优化结果进行了比较。结果正交实验结果为β-CD用量与挥发油比例为8:1,包合时间为3 h,包合温度为45℃;box-behnken实验结果为β-CD用量与挥发油比例为6:1,包合时间为3.4 h,包合温度为45℃。正交实验所需实验次数少,相对简单。结论用两种方法来寻求薄荷油-β-环糊精包合物最佳制备工艺都是可行的,两者实验结果基本接近,正交实验所需实验次数少相对简单,但Box-behnken实验设计在进行非线性模式设计和研究中具有优势。

【Abstract】 AIM To make a rational choice between orthogonal test design and box-behnken design by a comparison between the two methods in the preparation process of pennyroyal-β-cyclodextrin inclusion compound.METHODS β-cyclodextrin inclusion compounds were prepared by saturated water solution method.Independent variables of the experimental design were the feed ratio(w/v ratio) of β-cyclodextrin and pennyroya(X1),the inclusion time(X2) and the inclusion temperature(X3).Pennyroyal-β-cyclodextrin recovery ratio and drug entrapment efficiency were investigated as the index and optimized by respective orthogonal test design and box-behnken design,the comparison of optimized results was expected to be helpful in the reasonable selection of formulation optimization method.RESULTS The optimizational conclusion could be inferred from the two methods.The design optimum conditions of orthogonal test were as follows: the feed ratio(w/v ratio) of β-cyclodextrin and pennyroyal was 6∶ 1,the inclusion time was 3 h and the inclusion temperature was 60 ℃.The optimum conditions of box-behnken design were the feed ratio(w/v ratio) of β-cyclodextrin and pennyroyal was 6∶ 1,the inclusion time was 3.67 h and the inclusion temperature maintained at 50 ℃.Second-order polynomial equations model was used to analyze interaction effects between the multivariate factors via box-behnken design.CONCLUSION The two methods are suitable for optimizing the preparation of inclusion compound and experimentel results are approximate,Orthogonal test design is relatively simple and needs a few experiments.but box-behnken design predominates in the application of design and investigation in non-linearity model.
