川芎药材HPLC指纹图谱及阿魏酸测定研究
HPLC fingerprint of Chuanxiong Rhizoma and the determination of ferulic acid
【摘要】 目的建立川芎药材的高效液相色谱(HPLC)指纹图谱并测定阿魏酸。方法川芎的甲醇提取物采用RP-HPLC法,以Kromasil C18(4.6 mm×150 mm5,μm)柱为分析柱,甲醇-1%醋酸溶液梯度洗脱,体积流量0.8 mL/min,检测波长280 nm,柱温30℃,进样量10μL。结果以阿魏酸为参照峰,确定了8个共有特征峰,建立了10批川芎HPLC指纹图谱共有模式,对样品间的相似度进行了计算,并定量测定了样品中阿魏酸。结论所建立的HPLC指纹图谱及定量分析均具有较好的重复性,有助于川芎药材的质量评价。
【Abstract】 AIM To establish the HPLC fingerprints and determine ferulic acid of Chuanxiong Rhizoma.METHODS The methanlic extract of Chuanxiong Rhizoma was analysed by RP-HPLC.The Kromasil C18(4.6 mm×150 mm,5 μm) column was used,with methanol-1% acetic acid solution as mobile phase and gradient elution.The detective wavelength was set at 280 nm,the flow rate was 0.8 mL/min,and the column temperature was 30 ℃.RESULTS Ferulic acid as reference,the common model of HPLC fingerprint was stablished and 8 common peaks were identified.The similarities of different samples were compared.CONCLUSION The method of HPLC fingerprint and quantitative analysis is accurate,and it can be helpful in the quality control of Chuanxiong Rhizoma.
