槲皮素长循环纳米脂质体的小鼠口服吸收研究

【摘要】 目的制备槲皮素长循环纳米脂质体(QUE-LCL),并考察其理化性质及小鼠口服吸收特性。方法以山嵛酸甘油酯和聚山梨酯-80为载体材料,采用乳化蒸发-低温固化法,以正交实验法优化的处方与制备工艺制备槲皮素长循环纳米脂质体;透射电镜观察其微观形态,并测定其粒径分布;建立高效液相色谱法测定纳米脂质体和胃肠道内容物及粪样混合物中槲皮素含量。结果槲皮素长循环纳米脂质体为球状或类球状,平均粒径为172.63 nm,包封率为91.77%,其小鼠体内吸收优于槲皮素原料药和槲皮素普通脂质体。结论乳化蒸发-低温固化法适于制备槲皮素长循环纳米脂质体,以正交优化法制备的制剂促进小鼠对槲皮素的口服吸收。

【Abstract】 Objective To prepare long circulating nano-liposomes of quercetin(QUE-LCL) and investigate the physical and chemical properties and oral absorption in mice. Methods The QUE-LCL was prepared by the emulsified evaporation-low temperature solidification method with Glyceryl behenate(ATO) and Tween 80 as carriers and the optimum formulation selected by orthogonal test.Its morphology was examined by transmission electron microscope(TEM) and the particle diameter distribution was detected.The quereetin determination in nanoparticles and the mixture of gastrointestinal contents and feces by HPLC were established. Results QUE-LCL was spherical shape with mean diameter of 172.63 nm and entrapment efficiency was 91.77%.The oral absorption of QUE-LCL in mice was better than that of quercetin suspension and common liposomes. Conclusion The emulsified evaporation-low temperature solidification is suitable for preparing QUE-LCL,and the preparation of which made by orthogonal test promotes the oral absorption of quercetin in mice.
