RRM1表达与宫颈鳞癌新辅助化疗敏感性的关系
Expression of RRM1 in cervical squamous cell carcinoma and its relationship with the chemo-sensitivity to NACT
【摘要】 目的:研究核糖核苷酸还原酶M1(RRM1)在宫颈鳞癌中的表达及与新辅助化疗敏感性的关系。方法:运用免疫组织化学PV-6000二步法,检测40例接受新辅助化疗的宫颈鳞癌患者中RRM1的表达,与临床病理参数及与新辅助化疗敏感性的关系。结果:(1)新辅助化疗前,RRM1基因的表达率为40.0%(16/40),低分化者RRM1表达较中高分化者高(χ2=5.625,P=0.018),RRM1的表达与患者的绝经状态、高危型HPV感染、FIGO分期、淋巴结转移无关;(2)新辅助化疗后耐药组RRM1基因的表达率为87.5%(14/16),高于新辅助化疗前(56.2%),差异有统计学意义(χ2=4.038,P=0.044);(3)新辅助化疗前RRM1阳性组的化疗有效率为37.5%(6/16),RRM1阴性组的有效率为75.0%(18/24),差异有统计学意义(χ2=5.625,P=0.018)。结论:RRM1基因的表达与组织学分级有关,新辅助化疗可能上调RRM1基因的表达水平,RRM1有望成为预测宫颈癌新辅助化疗疗效的指标。
【Abstract】 Objective:To study the expression of ribonucleotide reductase M1(RRM1)and the relationship with the chemo-sensitivity to NACT(new adjuvant chemotherapy treatment) in cervical squamous cell carcinoma.Method:Using the immunohistochemistry PV-6000,we comparatively studied the expression of RRM1,the relationship among different clinical and biological features and the relationship with the chemo-sensitivity to NACT on 40 cases of cervical squamous cell who accepted NACT.Result:(1)Before NACT,the positive rate of RRM1 in cervical squamous cell carcinoma was 40.0%(16/40),the expression of RRM1 in lower differentiation cases was higher than in higher differentiation cases(χ2=5.625,P=0.018),the expression of RRM1 has no relationship with menopausal state,HPV infection,FIGO stage and lymph node metastasis.(2)After NACT,the expression of RRM1(87.5%)in drug resistance group was higher than before(56.2%).The difference was statistically significant(χ2=4.038,P=0.044).(3)Before NACT,RRM1 positive group of chemotherapy response rate was 37.5%(6/16),RRM1 negative effective rate was 75.0%(18/24),the difference was statistically significant(χ2=5.625,P=0.018).Conclusions:The expression of RRM1 was relavant to differentiation,NACT may raise the level of RRM1 expression.So RRM1 should be further investigated as the predictive markers for the chemo-sensitivity to NACT in cervical squamous cell carcinoma.
