金氏痔痛贴镇痛作用及其机制
Research of the analgestic effect of Jin’s hemorrhoid patch and its mechanism
【摘要】 目的:观察并研究金氏痔痛贴的镇痛作用及其机制。方法:将动物随机分为6组,即正常对照组、赋形剂组、止痛消炎软膏组及金氏痔痛贴大、中、小剂量组,分别进行药物干预7 d后。通过热板法及化学刺激法观察各组小鼠的镇痛作用;采用双抗体夹心法(E L I S A法)检测大鼠的血浆内皮素-1(endothelin-1,ET-1)、降钙素基因相关肽(calciton in gene-related peptide,CGRP)水平变化。结果:金氏痔痛贴对热传导及化学刺激引起的拟痛反应有明显的镇痛作用;且能使血浆ET-1水平显著下降;血浆CGRP水平显著升高,均具有统计学意义(P<0.01)。结论:金氏痔痛贴具有良好的镇痛功效,其缓解疼痛的机制可能是通过对血浆ET-1、CGRP水平的调整,从而缓解疼痛症状。
【Abstract】 Objective: To investigate the analgestic effect of Jin’s hemorrhoid patch.Methods: Animals were randomly divided into six groups,i.e.normal control group,Fuxing Ji group,ZTXY Ruan gao group and Jin’s hemorrhoid patch high-dose,middle-dose and low-dose groups.Jin’s hemorrhoid patch high-dose,middle-dose,and low-dose groups were treated respectively with 0.9 g crude drug /20 g(220 g),0.45 g crude drug/20 g(220 g) and 0.225 g crude drug/20 g(220 g),ZTXY Ruan gao group was treated with 0.45 g crude drug/20 g(220 g),Fuxing Ji group was treated with 0.45 g stroma/20 g(220 g),and normal control group was treated with 0.45 g NS/20 g(220 g).All groups were treated for 7 successive days.The analgesic effect of Jin’s hemorrhoid patch was tested by observation of the mouse’s reaction to the pain stimulated by heat conduction and chemical stimulus and plasma Calciton in gene-related peptide(CGRP),endothelin-1(ET-1)levels in contrast were investigated by ELISA assay.Results: Jin’s hemorrhoid patch could relieve the pain stimulated by heat conduction and chemical stimulus.Furthermore,it also could drop plasma ET-1 level and increase plasma level of CGRP significantly in rats,all had statistical significance(P <0.01).Conclusion:Jin’s hemorrhoid patch has strong analgesic effect which maybe through CGRP and ET-1 levels adjustment.
