氢氧化铝佐剂的过敏性反应

【摘要】 目的研究氢氧化铝佐剂的过敏性反应。方法取豚鼠42只,随机分为7组:A(lOH)31.5 mg/ml组、A(lOH)34 mg/ml组、A(lOH)3 7 mg/ml组、A(lOH)3 10 mg/ml组、A(lOH)3 13 mg/ml组、阴性对照(生理盐水)组和阳性对照(1%BSA)组,致敏,12 d后,分别经豚鼠静脉注射各浓度的A(lOH)3、生理盐水和1%BSA,观察各组豚鼠的反应及相关症状出现和消失的时间。另取豚鼠24只,随机分为4组:A(lOH)3 1.5 mg/ml组、A(lOH)3 4 mg/ml组、阴性对照(生理盐水)组和阳性对照(1%BSA)组,经豚鼠背部皮肤分别皮内注射抗血清,48 h后,各组分别静脉注射1.5 mg/ml、4 mg/ml的A(lOH)3、生理盐水和1%BSA+1%伊文思蓝染料(v/v=1∶1),进行激发;30 min后,测量豚鼠皮肤内层的蓝色斑点大小。结果全身主动过敏试验中,氢氧化铝浓度在4 mg/ml以下,豚鼠无过敏反应,7和10 mg/ml时为过敏反应强阳性,13 mg/ml时为过敏反应极强阳性。皮肤被动过敏试验中,A(lOH)3 1.5和4 mg/ml组豚鼠均未出现过敏反应。结论氢氧化铝浓度在4 mg/ml以下无过敏反应,较安全可靠。

【Abstract】 Objective To study the hypersusceptibility of aluminum hydroxide adjuvant.Methods Forty-two guinea pigs were divided into seven groups and sensitized then,12 d later,injected i.v.with Al(OH)3 1.5 mg / ml,Al(OH)3 4 mg / ml,Al(OH)3 7 mg / ml,Al(OH)3 10 mg / ml,Al(OH)3 13 mg / ml,physiological saline(negative control) and 1% BSA(positive control) respectively,and observed for reaction as well as the time of appearance and disappearance of the symptoms.Another 24 guinea pigs were divided into four groups and injected i.d.with antisera then,48 h later,injected i.v.with Al(OH)3 1.5 mg / ml,Al(OH)3 4 mg / ml,physiological saline and 1% BSA + 1% Evans blue(v / v = 1 ∶ 1) respectively for excitation.The sizes of blue spots on inner layers of guinea pigs were measured 30 min after excitation.Results Systemic active sensitization assay(ASA) showed that the Al(OH)3 at a concentration of less than 4 mg / ml caused no anaphylactic reaction in guinea pigs,while that at 7 ~ 10 mg / ml caused strong positive and that at 13 mg / ml caused extremely strong positive reactions.However,in passive cutaneous anaphylaxis(PCA) test,neither of Al(OH)3 at concentrations of 1.5 and 4 mg / ml caused anaphylactic reaction.Conclusion The Al(OH)3 at a concentration of less than 4 mg / ml caused no anaphylactic reaction,which was safe and reliable.
