蛹虫草子实体多糖的分离纯化

【摘要】 目的:研究人工培养蛹虫草子实体多糖的分离纯化方法。方法:多糖经脱色、醇析、除蛋白及乙醇分级沉淀,采用Seph-adex G100柱层析纯化。结果:60℃脱色9h脱色率达70.72%。醇析最优条件为:无水乙醇,24h,4倍乙醇。Sevag法抽提30min重复6次除蛋白率达84.09%;抽提1h重复3次、6次除蛋白率分别为81.14%、84.47%,多糖损失率分别为27.03%、43.16%。木瓜蛋白酶除蛋白最优条件为70℃,1:10,2 h,除蛋白率为25.67%。粗多糖经乙醇分级沉淀得2种粗多糖成分,分别经Sepha-dex G100柱层析可进一步纯化为3种较纯多糖成分。结论:采用乙醇分级沉淀及Sephadex G100柱层析纯化粗多糖可得到3种较纯多糖成分,纯化效果较好。

【Abstract】 Objective:This essay studied removing impurities methods of polysaccharides from fruiting body of cultured Cordyceps militaris.Method:Polysaccharides were separated with Sephadex G100 column chromatography after decoloured with 201×7 resin,precipitated with alcohol with orthogonal tests,deproteinized with Sevag and papain and preliminary separated with alcohol.Result:The rate of decolouration was 70.72% when decoloured 9h at 60 ℃.The optimal alcohol precipitation condition was using 4 times of absolute alcohol treating 24 hours.The deproteinization rate by Sevag was 84.09%when sample was extracted 30minutes repeated 6 times,that was 81.14% 84.47%separately when sample was extracted 1 hour and repeated 3 and 6 times respectively.Polysaccharides loss rate was 27.03%and 43.16%,respectively.The optal was 70 ℃,1:10,2 h when deproteinized with papain and deproteinization rate was 25.67%.Crude polysaccharides were separated into two components after alcohol separating precipitation,and were separated into three components with Sephadex G100 column chromatography.Conclusion:The purities of the polysaccharides were quite high after removed impurities and separated with Sephadex G100 column chromatography.
