谷胱甘肽对甲醛所致Hela细胞毒性的影响

【摘要】 为研究还原型谷胱甘肽(GSH)对甲醛所致Hela细胞毒性的影响,采用体外染毒方式,应用KCl-SDS法和MTT法分别检测GSH对甲醛引起Hela细胞DNA-蛋白质交联(DPC)和细胞活性的影响。结果表明,250μmol.L-1浓度下,DNA-蛋白质交联实验中甲醛染毒组所致的DPC显著高于对照组(p<0.01),GSH+甲醛组所致的DPC不但显著高于对照组(p<0.01),还显著高于甲醛染毒组(p<0.01),MTT实验中甲醛染毒组的细胞活性显著低于对照组(p<0.01),GSH+甲醛组所致的细胞活性不但显著低于对照组(p<0.01),还显著低于甲醛染毒组(p<0.01)。上述结果表明,GSH单独作用对Hela细胞无明显毒性作用,但是对甲醛所致毒性作用有显著促进作用。从而提出假设,在甲醛致白血病的过程中,GSH可能与甲醛结合形成活性状态,介导甲醛的毒性作用。

【Abstract】 The influence of glutathione(GSH) on toxicity of Hela cells induced by formaldehyde was studied.Hela cells were exposed to 250 μmol·L-1 formaldehyde,glutathione,the mixture of GSH and FA and a control,then KCl-SDS and MTT methods were used to determine DNA-protein crosslinks(DPC) and the activity of cells respectively.The results show that 250 μmol·L-1 formaldehyde induced significant DPC formation and decreased cell activity compared with the control group(p<0.01).At the same time,the mixture group could also induce significant DPC formation and decreased cell activity compared with the FA group(p<0.01).The results suggest that GSH did less harm to Hela cells.While GSH can enhance the toxicity of formaldehyde.Therefore,it can be proposed that in the process of leukemia induced by formaldehyde,glutathione could react with formaldehyde to affect the instant tissue.
