杜仲醇提物和水提物对小鼠免疫性肝损伤保护作用的研究

【摘要】 ①目的研究杜仲醇提物和水提物对卡介苗-脂多糖(BCG-LPS)所致小鼠免疫性肝损伤的保护作用。②方法小鼠90只,雌雄各半,随机分为9组,每组10只,即正常对照组、模型对照组、阳性对照组、醇提组和水提组的低、中、高剂量组(40.958、1.90、163.80g生药/kg)。观察杜仲提取物对小鼠肝脏组织病理学的影响。各组每天灌胃,给药容量为20mL/(kg.d),连续10天。正常组和模型组给予蒸馏水,阳性组给予联苯双酯0.60g/kg,醇提和水提组分别给予杜仲醇提物或水提物163.80、81.90、40.95g生药/kg。采用BCG-LPS造成小鼠急性肝损伤1,0天后取血检测ALT、AST、SOD的活性和MDA、GSH的水平。③结果杜仲醇提物和水提物能明显降低BCG-LPS所致的肝损伤小鼠肝脾指数的升高(P<0.05)和血清中ALT、AST活性的升高(P<0.05),亦能降低肝组织中MDA的水平,增加肝组织中SOD的活性和GSH-PX的水平(P<0.05)。肝脏组织病理学也显示联苯双酯和杜仲提取物能明显的对抗肝细胞的损伤,但杜仲水提物不如醇提物(P<0.05)。④结论杜仲醇提物具有显著的抗免疫性肝损伤的作用。

【Abstract】 Objective To evaluate the protective effects of the alcohol extract and water extract from Eucommia ulmoides on immunological liver-injury in mice induced by Bacille-Calmette-Guerin and lipopolysaccharide(BCG-LPS).Methods Mice were randomly divided into 9groups and 10 mice in each group,which were the normal group,model group,positive control group,3 dose alcohol extract group and 3 dose water extract group,6 test groups were given the ethanol or water extract at the dose of 40.95,81.90,163.80g/kg body weight,i.g once a day for 10 days,respectively,while positive group with bifendate(DDB)of 0.60g/kg b.w.in the same manner,and normal control and model control group with the same volume of distilled water.Immunological liver-injury model in mice was induced by BCG-LPS.The activities of ALT and AST in the serum,the content of MDA,and the activity of SOD,GSH in the liver cell were investigated.Results The ethanol extract from Eucommia ulmoides significantly decreased the level of ALT and AST in serum and the content of MDA in liver(P<0.01),but improved SOD and GSH activities(P<0.01)in the liver.Histologic examination indicated that most of liver cells were damaged after the treatment with BCG-LPS,but bifendate(DDB)and the extract from Eucommia ulmoides could significantly decreased the liver damage,but the effects of water extract were lower than the ethanol extract(P<0.05).Conclusion The ethanol extract from Eucommia ulmoides could significantiy resist the immunological liver-injury.
