几种不同种源远志等位酶与可溶性蛋白鉴别研究

【摘要】 目的:利用等位酶与可溶性蛋白凝胶电泳技术探讨远志不同种源的鉴别与亲缘关系。方法:采用聚丙烯酰胺凝胶电泳(PAGE)技术分析不同种源远志苹果酸酶、过氧化物酶、酯酶与可溶性蛋白谱带,采用NASYS软件对电泳谱带进行聚类分析。结果:远志可溶性蛋白与等位酶聚丙烯酰胺凝胶电泳图谱能够鉴别不同种源的远志,9份不同种源远志可被聚为三种类型,遗传距离0.1054~1.2040,除卵叶远志外,其余8份远志遗传距离0.1054~0.6931,卵叶远志与远志间的遗传距离明显高于远志间的遗传距离,地理位置相近的远志遗传距离较小,亲缘关系较近。结论:利用等位酶与可溶性蛋白凝胶电泳技术可为不同种源远志的鉴定与划分提供参考。

【Abstract】 Objective:Study on the identification and genetic relationship of Radix Polygalae from different provenances by allozyme and soluble protein gel electrophoresis technology. Methods:To analyse the bands of malic enzyme,peroxidase,esterase and soluble protein of Radix Polygalae from different provenances by polyacrylamide gel electrophoresis,and use NASYS software to process cluster analysis on these electrophoresis bands. Results:The allozyme and soluble protein bands of polyacrylamide gel electrophoresis can identify Radix Polygalae from different provenances,and nine different provenances can be clustered three classes,their genetic distance is between 0.1054 and 1.2040.Except P.sibirica L.,the others genetic distance is in 0.1054~0.6931,and there is apparently higher genetic distance both P.tenuifolia Willd.and P.sibirica L.than that among the populations of P.tenuifolia Willd.,and the geographically closer provenances have a lower genetic distance and a higher genetic relationship.Conclusion:It can provide reference for identifying and distinguishing Radix Polygalae from different provenances to use allozyme and soluble protein gel electrophoresis technology.
