葱白提取物纳米乳对急性心肌缺血兔血清C-反应蛋白、超氧化物歧化酶、丙二醛水平的影响
Influences of Fistular Onion Stalk Extract Nanoemulsion on serum CRP,SOD,MDA levels in acute myocardial ischemia Rabbit
【摘要】 目的观察葱白提取物不同制剂对急性心肌缺血兔血清C-反应蛋白(CRP)、超氧化物歧化酶(SOD)和丙二醛(MDA)的影响,评价葱白提取物纳米乳口腔喷雾剂治疗急性心肌缺血的适宜性。方法选用体重为2.5-3.0kg的健康雄性日本大耳白兔48只,随机分为假手术组、空白模型组、硝酸甘油组、葱白提取物组、葱白纳米乳组和葱白纳米乳原位凝胶组,每组8只。制备急性心肌缺血模型,术后及时给药。给药后观察3h,打开腹腔,腹主动脉取血,测定血清CRP、SOD、MDA的含量。结果葱白提取物组、葱白纳米乳组、葱白纳米乳凝胶组均能降低血清中MDA、升高SOD水平(均P<0.05),且不同制剂间没有显著性差异;葱白提取物及其两种纳米制剂均能显著降低CRP水平(均P<0.05),而葱白提取物纳米乳凝胶组降低CRP的水平显著优于硝酸甘油组(P<0.05)和葱白提取物组(P<0.05)。结论葱白提取物纳米乳原位凝胶在急性心肌缺血早期可以降低CRP水平,但是慢性的氧化损伤没有疗效优势,说明纳米乳原位凝胶可以用于急性心肌缺血治疗,值得深入研究。
【Abstract】 Objective To observe the effects of different preparations of Fistular Onion Stalk extract on serum C-reactive protein(CRP),malondialdehyde(MDA) and superoxide dismutase(SOD) in acute myocardial ischemia rabbits,and to evaluate the suitability of nanoemulsion Fistular Onion Stalk extract oral spray for treatment for acute myocardial ischemia.Methods 48 healthy male Japanese white rabbits weighing 2.5kg~3.0kg were randomly divided into sham operation group(n=8),the blank model group(n=8),nitroglycerin group(n=8),Fistular Onion Stalk extraction group(n=8),Fistular Onion Stalk extract nanoemulsion group(n=8) and Fistular Onion Stalk extract nanoemulsion gel in situ group(n=8).Rabbits were induced acute myocardial ischemia with timely medication administration after the operation,the sham operation group and model group were given saline(0~2mL/kg) sublingually,sublingual nitroglycerin group given nitroglycerin suspension(1mg·kg-1,5mg·ml-1).Three Fistular Onion Stalk extracts and dose sublingual formulation of nanoemulsion 20mg/kg,100mg/mL.Observed after administration for 3h,opened the abdominal cavity,pumped abdominal aortic blood,determined serum CRP,SOD,MDA contents.Results Fistular Onion Stalk extraction group,Fistular Onion Stalk extract nanoemulsion group,Fistular Onion Stalk extract nanoemulsion gel in situ group could reduce serum MDA,Increased SOD levels(P<0.05).Fistular Onion Stalk extract and its nano-emulsion,nanoemulsion gel in situ can resist oxidative damage,there was no significant difference between preparations.Fistular Onion Stalk extract nanoemulsion gel group could significantly lower CRP levels than nitroglycerin group(P<0.05) and Fistular Onion Stalk extract group(P<0.05).Conclusion Fistular Onion Stalk extract gel nanoemulsion in situ can reduce CRP levels in early acute myocardial ischemia,but there is no advantage of treating chronic oxidative damage,nanoemulsion gel in situ can be used to treat acute myocardial ischemia,it is worthy of further study.
