一株煤矿地下水硫酸盐还原细菌的分离、鉴定及性质研究
Isolation and identification of a SRB strain from coal mine groundwater and its characteristics analysis
【摘要】 从煤矿井下700 m深处含水层的钻孔涌水中分离获得一株硫酸盐还原细菌GY-2,该菌株革兰氏反应阴性,有芽孢,菌体棒状,能运动,氧化酶阴性;通过16S rRNA系统发育学分析,确定菌株GY-2为滴状脱硫肠状菌(Desulfotomaculum guttoideum)。GY-2菌株为兼性厌氧菌,在含有Fe2+的条件下生长良好;该菌株生长的pH值范围介于6.0~9.0之间,最适pH值为7.0~8.5,生长温度范围为20~45℃,最适温度为30~40℃;该菌株对盐度的耐受力较弱,生长的最适盐度为0.05%~1.0%。
【Abstract】 A sulfate-reducing bacteria(SRB) strain named GY-2 was isolated from the groundwater in a 700 m deep aquifer at a coal mine.The cells of strain GY-2 were found to be Gram-negative,spore-forming,rod-shaped,motile and oxidase-negative.Phylogenetic analysis based on 16S rRNA sequence identified the strain GY-2 as Desulfotomaculum guttoideum.Strain GY-2 is a facultative anaerobe and grows well as Fe2+ is presented.The pH range for strain GY-2 growth is 6.0-9.0(optimum pH value is 7.0-8.5) and the temperature range is 20-45 ℃(optimum temperature is 30-40 ℃).The salinity tolerance of strain GY-2 is poor and the optimum salinity range is 0.05%-1.0%.
