【摘要】 采用离子对色谱法测定细胞内α-酮戊二酸含量。色谱柱为Platisil C18柱,流动相为0.5%(NH4)2HPO4-5mmol.L-1四丁基氢氧化铵,H3PO4调pH值为4.0,检测波长为214 nm。结果表明,α-酮戊二酸与其它代谢物的分离效果较好,方法的线性相关系数为0.9996,相对标准偏差为0.38%,平均回收率为99.38%。更多还原

【Abstract】 An ion-pair chromatography method was established for the determination of content of α-ketoglutarate in E.coli BL21.A Platisil C18 column was used with the mobile phase of 0.5%(NH4)2HPO4-5 mmol·L-1 tetrabutylammonium hydroxide(pH value was adjusted by H3PO4 to 4.0) at the detection wavelength of 214 nm.The results showed that α-ketoglutarate was well separated from other metabolics.Its linear correlation coefficient was 0.9996,the relative standard deviation was 0.38%,the average recovery was 99.38%.更多还原
