草鱼cyclin G1基因的克隆、鉴定及适应性进化分析

【摘要】 本研究从草鱼(Ctenopharyngodon idellus)肝肾cDNA文库中克隆到草鱼cyclin G1基因。测序结果表明,草鱼cyclin G1全长cDNA为2118bp,使用ORF finder推译其开放阅读框为第256bp~第1155bp,编码299个aa,SMART在线软件预测其中N端第57个aa~第143个aa为细胞周期蛋白盒。同源性比对分析显示cyclin G1在鱼类中同源性极高,其中草鱼cyclin G1与斑马鱼的同源性为90%。通过PAML软件的密码子替换模型进行适应性进化分析,结果表明cyclin G1编码蛋白的26A、171A和225K氨基酸位点受到正选择作用。本研究为进一步了解草鱼cyclin G1结构功能与分子演化打下基础。

【Abstract】 In this research,we cloned cyclin G1 gene from the liver and kideny cDNA library in grass crap.The result of sequencing showed that the full length of cDNA of grass crap cyclin G1 was 2 118 bp,and the open reading frame was 256th~1 155th bp predicted by ORF finder which could code a 299 amino acid polypeptide.The analysis result of SMART on-line software showed that N-lobe the 57th~143th aa of grass carp cyclin G1 called cyclin box was a highly conserved region.Homology alignment results showed that cyclin G1 was highly homologous in fishes,grass carp cyclin G1 shared the homology of 90% with zebrafish.The adaptive evolutionary analysis on cyclin G1 protein studying by codon substitution model in PAML software indicated that amino acid sites of 26A,171A and 225K were suffered the positive selection effect.The research lays the foundation for further studying the structure,function and molecular evolution of grass carp cyclin G1.
