触须式聚合物开管柱的电色谱性能表征
Electricity Chromatographic Performance Characterization of Tentacle-Type Polymeric Open-Tubular Column
【摘要】 采用共价键合方法,制备了支化聚乙烯亚胺修饰的触须式聚合物开管柱,并对其电色谱性能展开了详细研究。开管柱的电渗流强度随着乙腈含量的升高而降低,随着流动相中缓冲液盐浓度的升高而减小。当流动相pH值低于7.5时,开管柱可获得反向电渗流;电渗流EOF日内、日间和周内的测定相对标准偏差RSD在0.8%—2.6%之间,柱间RSD为1.6%(n=4),批间RSD为4.8%(n=3),重现性较好。蛋白质在该柱上得到成功分离,没有严重拖尾。
【Abstract】 The tentacle-type polymeric open-tubular column that was modified by branched polyethyleneimine(PEI) was prepared by covalently bonded method,and the electrochromatography performance of which was detailedly investigated.The electroosmotic flow(EOF) strength of open-tubular column decreased along with increase of ACN and buffer salt concentration in mobile phase.When pH value of mobile phase was less than 7.5,the open-tubular column got the reversed EOF,and the days,daytime and weeks RSDs for EOF were in the range of 0.8%—2.6%,and between the columns and columns RSD was 1.6%(n=4),while batch and batch RSD was 4.8%(n=3),that showed the good repeatability.The favorable separation of proteins was successfully obtained without obvious peak tailing.
