附子理中丸对脾阳虚证大鼠骨骼肌能荷影响的实验研究

【摘要】 目的:观察附子理中丸对脾阳虚证大鼠骨骼肌能荷的影响。方法:18窝新生大鼠按同窝匹配、体重均衡原则随机分9组,前5组不切除甲状腺作为获得性脾阳虚,后4组切除甲状腺作为发育性脾阳虚。d1切除部分甲状腺。d42切除肩胛骨间棕色脂肪,d56高脂饲养和d70用药防治。d98取骨骼肌,高效液相色谱测量能荷。t检验比较组间效能差别。结果:获得性脾阳虚大鼠中,阳虚组能荷值比正常组降低14.2%(P<0.01),脾阳虚组能荷值比阳虚组降低16.0%(P<0.01),凉治组能荷值比脾阳虚组升高5.8%(P<0.01),温治组能荷值比脾阳虚组降低9.6%(P<0.01)。发育性脾阳虚大鼠中,阳虚组能荷值比正常组升高45.6%(P<0.01),脾阳虚组能荷值比阳虚组降低23.9%(P<0.01),凉治组能荷值比脾阳虚组升高6.8%(P<0.01),温治组能荷值比脾阳虚组降低16.8%(P<0.01)。与获得性脾阳虚比较,发育性脾阳虚阳虚组、脾阳虚组、凉治组和温治组的能荷值分别升高69.7%、53.8%、55.4%和41.7%(P<0.01)。结论:降低骨骼肌能荷是附子理中丸增强发育性和获得性脾阳虚大鼠产热潜能的机制之一。

【Abstract】 Objective:To observe the effects of aconitum lizhong pills on the levels of energy charge in the skeletal muscle of rats with spleen yang deficiency syndrome.Method:Eighteen littermates,each litter was bred sequentially and random divided into 9 groups.Five groups were not carried out thyroidectomy,the other four groups did.Thyroidectomy in hypothyroidism littermates were carried out with 131iodine to destroy partly thyroidal tissue on d1 after birth,the interscapular brown adipose tissue was removed on d42,littermates were fed with more cholesterol ester and triglyceride from d56,then administered intragastrically a decoction of medical herbs to promote or inhibit chronic thermogenesis from d70.The levels of energy charge in skeletal muscle was quantified by HPLC at d98.The potency data derives from the primary data.A t-test was used to evaluate the change between groups,P<0.05 meaned statistic significance. Result:In the euthyroid rats,compared with the control group,the levels of energy charge in skeletal muscle was decreased by 14.2%(P<0.01) in model rats(rats with yang deficiency syndrome).The level in fat-diet rats(rats with spleen yang deficiency syndrome)was decreased 16.0%(P<0.01) than that in the model rats.The rats treated with huanglian jiedu decoction increased the levels by 5.8%(P<0.01) and the rats treated with aconitum lizhong pills decreased by 9.6%(P<0.01) than that in the fat-diet rats.In hypothyroid rats,compared with the control group,the levels of energy charge in skeletal muscle increased by 45.6%(P<0.01) in model rats;the level in fat-diet rats decreased 23.9%(P<0.01) than that in the model rats;the rats treated with huanglian jiedu decoction increased the levels by 6.8%(P<0.01) and the rats treated with aconitum lizhong pills decreased by 16.8%(P<0.01) than that in the fat-diet rats.The levels potency of energy charge in the groups with hypothyroid were higher than that in the design -mated groups with euthyroid(P<0.01),by 69.7%in model group,53.8%in fat-diet group,55.4%in huanglian jiedu decoction group and 41.7%in aconitum lizhong pills group.Conclusion:By decreasing the energy charge,at least in part,aconitum lizhong pills can enhance significantly the thermogenesis of skeletal muscle in the rats with spleen yang deficiency syndrome,more effective potency in the rats with hypothyroid than those with euthyroid.
