【摘要】 目的:观察脾阳虚证大鼠胃生长素(Ghrelin)改变的特征,以及温阳健脾方附子理中汤对脾阳虚证Ghrelin的改善情况,以探讨Ghrelin在脾阳虚证发病机制中的作用和附子理中汤干预脾阳虚证可能的药效环节。方法:实验动物新生大鼠饲养6周后按体重随机均衡分为4组,每组16只。阳虚组手术切除BAT,脾阳虚组复合高脂饲料与寒冷刺激,中药治疗组给予附子理中汤,对照组不施加任何处理,4周后取材检测血清Ghrelin水平。结果:脾阳虚组和对照组相比,体重增长率明显下降(P<0.01);中药组和脾阳虚组相比,体重增长率上升(P<0.01)。脾阳虚组和对照组相比,血清Ghrelin含量降低明显(P<0.01);中药组和脾阳虚组相比,血清Ghrelin含量升高(P<0.01)。结论:实验结果提示,Ghrelin水平下调可能是脾阳虚证大鼠模型形成机制中的神经内分泌基础之一,而附子理中汤可通过上调Ghrelin水平来改善脾阳虚证,从而达到"健脾"和"温阳"的功效。更多还原

【Abstract】 Objective:To explore the function of Ghrelin in the pathogenesis of spleen yang deficiency syndrome and possible regulating mechanism of Ghrelin in rats with spleen yang deficiency syndrome after administrated by Aconitum Lizhong decoction by observing traits of Ghrelin change.Methods: The rats are divided into 4 groups,and each group including 16 cases.①control group: fed with ordinary food and water,d70-d98 ig.with 0.9% NaCl 10 ml/kg/d.②yang deficiency syndrome group: the same as the control group’s except for removing brown adipose tissue on d42.③spleen yang deficiency syndrome group: the same as the yang deficiency syndrome group’s except that fed with high fatty diet(including 83% ordinary diet,15% triglycerides,2% cholesterol) from d49 to d98 and placed in circumstance of 28 ℃ and 19℃ in turn.④Chinese herbal decoction group: the same as the spleen yang deficiency syndrome group’s except of ig.5ml /kg /d with Aconitum Lizhong decoction.Results: compared with the control group,rate of weight growth of the spleen yang deficiency group both drops evidently(P < 0.01);compared with the Chinese herbal decoction group,rate of weight growth of spleen yang deficiency group rises(P < 0.01);compared with the control group,level of serum Ghrelin of the spleen yang deficiency group drops evidently(P < 0.01),compared with the spleen yang deficiency group,level of serum Ghrelin of the Chinese herbal decoction group rises(P < 0.01).Conclusions: Down-regulating of level of serum Ghrelin is possible one of neuro-endocrine bases during the forming process of rat model with spleen yang deficiency syndrome;however,in the mechanism of improving symptoms of spleen yang deficiency syndrome.up-regulating level of serum Ghrelin is possible one of neuro-endocrine bases which have functions of refreshing spleen and warming yang.更多还原
