猪链球菌RAPD-PCR(RAPD)反应条件的优化
Optimization on the condition for RAPD-PCR of Streptococcus suis gene
【摘要】 研究猪链球菌随机扩增多态性DNA(RAPD)扩增条件。结果表明,当MgCl2浓度为2mmol/L,DNA模板为50ng,dNTP加入量为1.5μL/25μL时扩增效果最好;引物SBSBG07、SBSBG10、SBSBG17及SBSE03、SBSE05、SBSE11号扩增效果较好,即有6条引物可获得理想的扩增结果。
【Abstract】 Random amplified polymorphic DNA PCR were carried out to optimize and screene the best RAPD-PCR reaction system and reaction process of Streptococcus suis gene. The results were following: each 25 μL amplification reaction solution including 2 μL MgCl2, 50 ng template DNA and 1.5 μL dNTPs showed optimal amplified effects; 6 primers were screened suit for the RAPD-PCR of Streptococcus suis, that were SBS BG07, SBS BG10, SBS BG17, SBS E03, SBS E05 and SBS E11.
