注意缺陷多动障碍大鼠前额叶中Homer家族表达研究

【摘要】 目的探讨Homer家族在注意缺陷多动障碍(ADHD)动物模型幼年自发性高血压(SHR)大鼠与Wistar京都(WKY)大鼠前额叶中的表达差异情况,分析其与ADHD的关系。方法2006年12月至2007年7月在南京医科大学儿科医学研究所应用反转录-聚合酶链反应(RT-PCR)、免疫蛋白印迹技术(Western Blotting)检测SHR与WKY大鼠前额叶皮质中Homer1a、Homer1b/c、Homer2a/bmRNA和蛋白的表达情况。结果RT-PCR结果显示,SHR大鼠Homer1a、Homer2a/bmRNA表达量较WKY大鼠降低,差异有统计学意义(t=2.56,P<0.05;t=5.85,P<0.05),Homer1b/c在两组间差异无统计学意义(t=0.44,P>0.05)。Western Blotting结果显示,SHR大鼠Homer1a、Homer2a/b蛋白表达量较WKY大鼠降低,差异有统计学意义(t=2.76,P<0.05;t=3.40,P<0.05)。结论SHR大鼠前额叶皮质Homer1a、Homer2a/b表达较WKY大鼠显著降低,提示Homer1a、Homer2a/b可能与ADHD密切相关。

【Abstract】 Objective To investigate the expression of Homer on Attention-Deficit Hyperactivity Disorder (ADHD). Methods The total RNA and protein were extracted from prefrontal cortex (PFC) of SHR and WKY rats. Both RT-PCR technique and Western Blotting technique were used to detect the expression levels of different isoforms of Homer (Homer 1a, Homer 1b/c, Homer 2a/b) in PFC of SHR and WKY rats. Results (1) By RT-PCR detection, it was found that the expression levels of Homer 1a and Homer 2a/b gene were significantly lower in prefrontal cortex of SHR rats than those in WKY ones (t = 2.56,P < 0.05;t = 5.85,P < 0.05). There was no difference in the expression levels of Homer1b/c gene in prefrontal cortex between SHR and WKY rats (t = 0.44,P > 0.05). (2) By Western Blotting, it was found that the expression levels of Homer 1a and Homer 2a/b gene were significantly lower in prefrontal cortex and hippocampus of SHR rats than those in WKY rats( t = 2.76,P < 0.05; t = 3.40,P < 0.05). Conclusion Homer 1a and Homer 2a/b may play an important role in pathogenesis of ADHD.
