曲古抑菌素A对人胃癌SGC-7901细胞凋亡及其RASSF1A基因表达的影响
Effects of TSA on Apoptosis and Expression of RASSF1A Gene in Human Gastric Cancer SGC-7901 Cells
【摘要】 目的探讨组蛋白去乙酰化酶抑制剂曲古抑菌素A(TSA)对人胃癌SGC-7901细胞凋亡及其RASSF1A基因表达调控的影响。方法用0.1、0.3、1.0μmol/LTSA分别处理人胃癌SGC-7901细胞,流式细胞仪检测细胞凋亡率,RT-PCR、Western blot分别检测RASSF1AmRNA和蛋白表达水平。结果流式细胞仪分析表明,不同浓度TSA可诱导SGC-7901细胞凋亡,并呈浓度及时间依赖性(P<0.05)。TSA干预前SGC-7901细胞无RASSF1AmRNA及蛋白表达,而不同浓度TSA处理SGC-7901细胞后,RASSF1AmRNA及蛋白表达分别上调,呈浓度及时间依赖性(P<0.05)。结论TSA诱导SGC-7901细胞凋亡可能与RASSF1A基因异常乙酰化状态恢复导致基因重新表达有关。
【Abstract】 Objective To investigate the effects of the histone deacetylation inhibitor,trichostatin A (TSA) on the expression of RASSF1A gene in human gastric cancer cell line.Methods Gastric cancer SGC-7901 cells were treated with different concentration of TSA.The apoptosis of the cells were detected by flow cytometry.RT-PCR and Western blot were applied to detect the RASSF1A mRNA and protein expression levels,respectively.Results Flow cytometry analysis showed that the apoptosis rate in SGC-7901 cells went up significantly depended on the dose and time(P<0.05).The mRNA and protein expression of RASSF1A gene was not detected in gastric cancer SGC-7901 cells before treatment.After treated with TSA at different concentrations and time,mRNA and protein expression of RASSF1A gene increased in a dose-and time-dependent manner (P<0.05).Conclusion It is possible that TSA induced the apoptosis of gastric cancer SGC-7901 cells by eliminating the aberrant histone acetylation status of RASSF1A gene.
