细菌脂多糖在人牙龈成纤维细胞膜表面的定位研究
The Location of LPS on Human Gingival Fibroblasts Surface
【摘要】 目的研究利用荧光显微镜观察牙龈成纤维细胞膜表面的荧光表达,细菌脂多糖在人牙龈成纤维细胞膜表面的定位。方法组织块法培养人牙龈成纤维细胞,利用荧光显微镜观察牙龈成纤维细胞膜表面的荧光表达,同时观察mCD14抗体对脂多糖结合牙龈成纤维细胞膜表面的阻断。结果荧光显微镜观察结果表明,异硫氰酸荧光素标记脂多糖可直接结合在牙龈成纤维细胞的膜表面的受体,且随着脂多糖浓度的升高,细胞表面的荧光强度升高,mCD14抗体可以部分阻断脂多糖结合牙龈成纤维细胞膜表面。结论细菌脂多糖可以结合在牙龈成纤维细胞膜表面,mCD14可能是脂多糖在牙龈成纤维细胞膜表面的受体,在识别LPS的过程中起关键的作用。
【Abstract】 OBJECTIVE To Study the location of lipopolysaccharide(LPS) on human gingival fibroblasts(HGFs) surface.METHODS We observed the immunofluorescence of HGFs surface to study the LPS-binding on HGFs with a fluorescence microscope.RESULTS It was demonstrated that FITC-LPS could bind to the receptor of cells.FITC-LPS bound to HGFs more strongly with the increasing concentration of LPS.LPS binding could be partly blocked by preincubation of HGFs with anti-CD14 monoclonal antibody.CONCLUSIONS The results indicate that LPS can bind to HGFs and the modulation of HGFs function depend on mCD14.
